摘要
目的:研究虾青素(AST)预保护对脂多糖(LPS)诱导的小鼠急性肠道炎症的影响。方法:健康雄性昆明小鼠40只随机分为4组:对照组、虾青素组、LPS组和虾青素+LPS组。在腹腔注射生理盐水或LPS(1 mg/kg)前,各组连续15 d分别灌胃橄榄油或虾青素(50 mg/kg)。LPS注射3 h后处死小鼠,采集血液及十二指肠、空肠和回肠组织。取材后通过ELISA和荧光定量PCR检测各组血清及各肠段组织中TNF-α、IL-6和IL-1β等炎症因子的分泌量和mRNA的表达量,Western blot检测NF-κB p65蛋白表达量,并通过HE染色观察各个肠段组织病理形态学变化。结果:与LPS组相比,虾青素+LPS组小鼠血清和肠道组织中炎症因子的分泌和mRNA的表达量及NF-κB p65蛋白表达量均显著降低(P <0. 05),且小肠段黏膜损伤程度低,绒毛形态均匀完整。结论:虾青素可改善小鼠肠黏膜结构和功能,并且通过抑制NF-κB通路中NF-κB p65蛋白表达,降低机体中炎性因子TNF-α、IL-6和IL-1β的分泌来缓解LPS引起的急性肠道损伤。
Objective: To investigate the effects of astaxanthin(AST) treatment on acute intestinal inflammation induced by lipopolysaccharide(LPS) in mice.Methods: Forty healthy male Kunming mice were randomly allocated into four groups: control group,AST group,LPS group and AST + LPS group.AST and olive oil was administrated by oral gavage consecutively for 15 days before the saline or LPS(1 mg/kg) peritoneal injection.Blood,duodenum,jejunum and ileum tissues were collected at 3 h after LPS injection.The levels of inflammatory factors such as TNF-α,IL-6,and IL-1β in serum and intestinal tissues were measured by ELISA and fluorescence quantitative PCR,NF-κB p65 protein was measured by Western blot.H&E staining was used to observe the histopathological changes.Results: Compared with LPS group,the secretion and the relative expression of inflammatory factors,the same as the expression of NF-κB p65 protein were significantly reduced in serum and intestinal tissue of AST + LPS group(P < 0.05).In addition,the degree of intestinal mucosal damage was low,and the intestinal villi were uniform and intact.Conclusion: AST treatment can improve the structure and function of intestinal mucosa,and relieve acute intestinal injury by suppressing the secretion and relative expression of inflammatory cytokines(TNF-α,IL-6 and IL-1β),and inhibiting the expression of NF-κB p65 protein in NF-κB pathway in LPS-induced mice.
