摘要
为了利用东北大米资源,分离获得糖化大米用真菌。通过稀释涂布平板法,划线以及点种的方法从自然样品中分离出形态不同的霉菌,采用透明圈法初筛,利用糖化能力测定法复筛,并对具有糖化能力的菌株进行形态学和分子生物学的测定,筛选出有糖化作用的真菌作为大米糖化用的新菌株。共得到11株形态不同的霉菌,其中6株具有糖化能力,通过酶活力测定复筛得到2株高产糖化酶和淀粉酶的菌株T4和T6,其中T4的糖化力和液化力分别为479.1 U·g^(-1)、188.0 U·g^(-1),T6的糖化力和液化力分别为485.6 U·g^(-1)、177.3 U·g^(-1)。通过ITS测序鉴定T4与黄曲霉Aspergillus flavus、T6与米曲霉Aspergillus oryzae相似性最高。T6是一株具有应用潜力的大米用糖化菌株。
In order to utilize excellent rice resources in northeast China,saccharified rice fungi was separated.different shapes of fungi from natural samples with methods of dilution-plate,streaking and seeding was separated,then clarity circle method was used to preliminarily screen,saccharifying ability method to rescreen and the strains which had saccharifying ability were measured their morphology and molecular biology to screen a new strain.11 strains of different types of fungi were obtained,among them 6 had saccharifying ability.2 strains of high-yield glucoamylase and amylase were rescreened by measuring enzyme activity,named T4 and T6,respectively.The saccharifying power of T4 and T6 were 479.1 U·g^-1 and 485.6 U·g^-1,liquefying power of T4 and T6 were 188.0 U·g^-1 and 177.3 U·g^-1,respectively.T4 had the highest similarity with Aspergillus flavus,equally,T6 was similar as Aspergillus oryzae by using ITS sequencing identification.T6 was a strain which had saccharify rice with application potential.
