摘要
研究表明,黑变红枣枣皮类黑精的最大吸收波长为280nm,本试验以黑变红枣枣皮为原料,在单因素试验的基础上,通过响应面分析法优化水浴与细胞破碎联动对黑变红枣枣皮类黑精提取工艺并建立回归模型。结果表明:最佳提取工艺条件为水浴时间100min、水浴温度78℃、细胞破碎时间为53min、细胞破碎功率66%。在此工艺条件下,测得黑变红枣枣皮吸光度为1. 072。与理论值1. 031相比,相对误差约3. 9%。与常规水浴法和细胞破碎提取黑变红枣枣皮类黑精相比,联动水浴和细胞破碎提取效果更好。最佳提取级数为3级,提取率为91. 38%。
Study shows that the maximum absorption wavelength of melanin in blacken jujube peel melanoidin is 280nm.With black jujube peel as raw material,on the basis of single factor experiment,response surface methodology was used to optimize the extraction process of melanosperm from blacken jujube peel melanoidin by linkage water bath and cell breakage and establish regression model.The results showed that the optimum extraction conditions were water bath time of 100min,water bath temperature 78℃,cell fragmentation time of 53min,and cell fragmentation amplitude of 66%.Under this technological condition,the absorbance of black jujube peel was 1.072.Compared with the theoretical value of 1.031,the relative error was about 3.9%.Compared with the conventional water bath method and cell crushing method,the combined water bath and cell crushing extraction effect were better.The optimum extraction stage was 3 and the extraction rate was 91.38%.
