摘要
目的研究纯化后富含EPA和DHA的鱼油对实验性高脂血症大鼠血脂和脂质过氧化的影响。方法采用高脂饮食喂饲法建立高脂血症大鼠模型,将60只大鼠随机分为正常对照组、高脂模型组、月见草油组270 mg/(kg·d)、鱼油低剂量组330 mg/(kg·d)、中剂量组670 mg/(kg·d)、高剂量组1 330 mg/(kg·d);最后测定各组甘油三酯(TG)、血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、血清中丙二醛(MDA)和超氧化物歧化酶(SOD),并作病理学观察。结果鱼油的3个剂量均能降低高脂大鼠的TC、TG、LDL-C,仅高剂量组能升高HDL-C;鱼油高剂量组能显著抑制MDA含量上升,提高SOD活力,中剂量组疗效次之;镜下观察:正常组无异常,肝小叶结构清晰,肝细胞索由中央静脉向四周呈放射状排列;模型组肝小叶结构轮廓基本消失,肝细胞体积增大,部分水样变性,部分脂肪变性;鱼油低剂量组与模型组相似;中剂量组脂变程度减轻,再增加剂量,最轻的是高剂量组肝脏脂变程度,肝小叶周边的细胞内仅有一些细小的脂肪滴散布。结论随着剂量的增加,鱼油具有调节血脂代谢异常和抗脂质过氧化作用,提示高剂量鱼油可能对治疗高脂血症有一定的应用价值。
Objective To study the effect of fish oil that is rich of EPA and DHA after purification on blood-lipid and lipid peroxidation of experimental rats with hyperlipemia.Methods 60 rats were selected to establish the hyperlipidemia rat model by feeding high fat diets and were randomly divided into normal control group,high fat model group,evening primrose oil group 270 mg/(kg·d),low dose fish oil group of 330 mg/(kg·d),middle dose group of 670 mg/(kg·d)and high dose group of 1 330 mg/(kg·d).Triglycerides(TG),serum total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),serum malondialdehyde(MDA)and superoxide dismutase(SOD)were tested in the end and were observed in pathology.Results All three doses of fish oil could decrease the TC,TG,LDL-C of high fat rats.Only the high dose of fish oil could increase the HDL-C of rats.High dose fish oil could significantly inhibit the increase of MDA content and increase the activity of SOD.The effect of the middle dose group came the next.Microscopic observation showed that there was no abnormal structure in the normal group:the structure of the liver lobuli was clear,the liver cells around the central vein were in radial pattern,and in the model group,the hepatis structure outline basically disappeared with the volume of liver cells increasing,and hydropic degeneration and fatty degeneration occurred in certain areas.Low dose fish oil group and model group were similar.The degree of lipid changes decreased in the middle dose group.The low dose group of fish oil was similar to the model group;in the middle dose group,the degree of lipid change was alleviated and when the dosage increased,the variation degree of liver fat of the high dose group was minimal with only a small number of fat droplets scattering in the surrounding cells.Conclusion With the increase of dosage,fish oil has an effect on regulating blood-lipid metabolic disorder as well as preventing lipid peroxidation,which suggests that high dosage of fish oil may have certain application value in treating hyperlipemia.
