摘要
目的:通过研究长链非编码RNA(LncRNA)-LINC00152对胶质瘤细胞侵袭转移的影响,进一步探究LncRNA-LINC00152在胶质瘤发生发展中的作用机制。方法:收集胶质瘤标本(69例)及癌旁标本(69例),通过RT-PCR技术对LncRNA-LINC00152 mRNA的表达水平进行定量,同时分析LncRNA-LINC00152表达水平与患者临床病理特征的相关性。此外,小干扰RNA(siRNA)在人胶质瘤细胞系U118和U251中构建LncRNA-LINC00152稳定敲除的细胞系,并利用划痕实验及Transwell侵袭实验检测胶质瘤细胞的侵袭及迁移能力。同时利用免疫印迹技术,对TGF-β/Smad3信号通路相关蛋白表达水平进行检测。结果:胶质瘤组织中LncRNA-LINC00152 mRNA表达水平明显高于癌旁组织(P<0.05);利用siRNA抑制LncRNA-LINC00152后,U118和U251细胞侵袭及迁移能力明显受到抑制(P<0.05)。LncRNA-LINC00152敲除后,TGF-β/Smad3信号通路的蛋白表达被抑制(P<0.05)。结论:抑制胶质瘤细胞中的LncRNA-LINC00152表达可以抑制肿瘤细胞的侵袭及转移,其机制可能与LncRNA-LINC00152介导的TGF-β/Smad3信号通路有关。
Objective:To study the effect of long chain noncoding RNA(LncRNA)-LINC00152 on invasion and metastasis of glioma cells.To further explore the role of LINC00152 in the development of glioma.Methods:Clinical specimens of glioma(69 cases)and paracancerous specimens(69 cases)were collected.The expression level of LINC00152 was quantified by RT-PCR,and the correlation between LINC00152 expression and clinicopathological features was analyzed.In addition,small interfering RNA(siRNA)was used to construct LINC00152 stable knockout cell lines in human glioma cell lines U118 and U251,and the invasion and migration ability of glioma cells was detected by scratch test and Transwell invasion assay.At the same time,the expression level of TGF-β/Smad3 signal pathway related protein was detected by Western blotting.Results:The expression of LINC00152 mRNA in gliomas was significantly higher than that in paracancerous tissues(P<0.05).After the inhibition of LINC00152 by siRNA,the invasion and migration of U118 and U251 cells were significantly inhibited(P<0.05).After LINC00152 knockout,the protein expression of TGF-β/Smad3 signaling pathway was inhibited(P<0.05).Conclusion:Inhibiting the expression of LINC00152 in glioma cells can inhibit the invasion and metastasis of tumor cells.The mechanism may be related to the TGF-β/Smad3 signaling pathway mediated by LncRNA-LINC00152.
