摘要
目的探讨低情绪对雌性大鼠血清性激素作用及与多囊卵巢综合征(PCOS)的关系。方法抑郁模型制作参考Gronli的方法。大鼠单笼饲养,每日给予1~2种刺激,每种刺激不连续使用,持续4周。这些刺激包括禁食、禁水、斜笼、湿笼、持续光照和水平摇晃等,建立PCOS大鼠模型。将大鼠进行open-field法行为学评分,选出合格大鼠,随机分为正常健康组、轻度抑郁组、重度抑郁组,进行不同程度刺激的造模。使用血糖仪测定大鼠GLU,计算AUC、HOMA-IR及ISI,行阴道脱落物涂片、HE染色观察动情周期,用ELISA法检测FSH、LH、T及INS水平。结果与健康组相比,轻度抑郁组和重度抑郁组大鼠的空腹GLU、空腹INS和AUC、HOMA-IR指数、INS、LH、T、卵巢重量均有不同程度上升,ISI指数下降,体重及BMI值、FSH(P>0.05)无明显差异,无规律的动情周期,无排卵。结论低情绪可影响雌性大鼠体内血清性激素含量,并引发相关的多囊卵巢综合征的发病。
Objective To investigate the effect of low emotion on serum sex hormones in female rats and its relationship with polycystic ovary syndrome(PCOS).Methods The Depression model was made according to the method of Gronli.Rats are raised in single cages,giving 1~2 stimuli per day,each of which is not used continuously for 4 weeks.These stimuli include fasting,forbidden water,sloping cages,wet cages,continuous illumination and horizontal shaking.Then,establish the PCOS model of rats.The eligible rats were randomly divided into normal health group,mild depression group and severe depression Group by Open-field behavioral score.At the same time,the modeling of different degree of stimulation was carried out.The GLUl of rats was determined by blood glucose meter,and calculate AUC,HOMA-IR and ISI.Observe the emotional cycles by vaginal exfoliation smear under he staining.Detect the levels of FSH,LH,T and INS by ELISA.Results Compared with the health group,the fasting GLU,fasting INS and AUC,Homa-ir index,INS,LH,T and ovary weight of rats in mild depression group and severe depression group increased in different degrees,the ISI index decreased,and there was no significant difference between body weight and BMI value and FSH(P>0.05).Meanwhile,there is no regular emotional cycle and no ovulation.Conclusion Low mood can affect the content of serum sex hormones in female rats and cause the pathogenesis of polycystic ovary syndrome.
