丹防胶囊对免疫性肝纤维化大鼠肝组织IL-33和ST2表达的影响 被引量:5

摘要 目的:探讨丹防胶囊对免疫性肝纤维化大鼠肝组织白细胞介素33(IL-33)、肿瘤发生抑制蛋白2(ST2)表达的影响。方法:40只清洁级健康雄性SD大鼠按随机数字法平均分为模型组、阴性对照组、丹防组及阳性对照组,模型组、丹防组、阳性对照组大鼠每只给予腹腔注射0.5 mL猪血清制备大鼠免疫性肝纤维化模型,阴性对照组腹腔注射等量生理盐水;造模同时,丹防组予4.32 g/kg丹防胶囊灌胃,阳性对照组予0.54 g/kg复方鳖甲软肝片灌胃,模型组与阴性对照组予等量生理盐水灌胃(1次/d),共12周;12周末麻醉处死大鼠后行肝组织病理学检查,免疫组织化学法、Western Blot检测肝组织中IL-33、ST2蛋白表达,Q-PCR检测肝组织中IL-33 mRNA表达水平。结果:与阴性对照组比较,模型组肝纤维化明显,模型组和丹防组IL-33、ST2蛋白及IL-33 mRNA表达增加,差异有统计学意义(P<0.05);与模型组比较,丹防组、阳性对照组肝纤维化程度明显减轻,IL-33、ST2蛋白及IL-33mRNA表达降低,差异有统计学意义(P<0.05);与阳性对照组相比,丹防组肝纤维化减轻程度差异无统计学意义(P>0.05),但IL-33、ST2蛋白及IL-33mRNA表达增多,差异有统计学意义(P<0.05)。结论:丹防胶囊对大鼠免疫性肝纤维化有一定干预作用,其机制可能与降低肝纤维化大鼠肝组织IL-33及ST2的表达有关。 Objective: To explore the effects of Danfang capsule on the expression of Interleukin-33(IL-33) and Tumorigenicity 2 receptor(ST2) in the liver tissue of rats with immune hepatic fibrosis. Methods: Forty clean healthy male SD rats were evenly divided into model group, negative control group, Danfang group and positive control group according to the random number method. The model group, Danfang group and positive control group were intraperitoneally injected with 0.5 mL pig serum/rat to prepare the rat model of immune liver fibrosis, and the negative control group was intraperitoneally injected with the same amount of normal saline. At the same time, the Danfang group was given 4.32 g/kg Danfang capsule gavage, the positive control group was given 0.54 g/kg compound turtle shell soft liver tablets gavage, and the model group and the negative control group were given the same amount of normal saline gavage once a day for a total of 12 weeks. Liver histopathological examination was performed after the rats were anesthetized to death at the end of 12 weeks. The expression levels of IL-33 and ST2 proteins in liver tissues were detected by immunohistochemical method and Western Blot, and the expression levels of IL-33 mRNA in liver tissues were detected by Q-PCR. Results: Compared with the negative control group, liver fibrosis was significant in the model group, and the expressions of IL-33, ST2 protein and IL-33 mRNA increased in the model group and the Danfang group(P<0.05). Compared with the model group, the degree of liver fibrosis was significantly reduced in the Danfang group and the positive control group, and the expressions of IL-33, ST2 protein and IL-33 mRNA decreased(P<0.05). Compared with the positive control group, there was no significant difference in the degree of liver fibrosis reduction in the DanFang group(P>0.05), but the expression of IL-33, ST2 protein and IL-33 mRNA increased, and the difference was statistically significant(P<0.05). Conclusion: Danfang capsule has a certain intervention effect on immune liver fibrosis in rats, and the mechanism may be related to the inhibition of proliferation and activation of HSC by reducing the expression of IL-33 and ST2.
