摘要
目的探讨不同剂量的黄连解毒汤对SD大鼠粪便脂肪酸代谢的影响,为临床合理使用清热解毒药提供一定的参考资料。方法将健康SD大鼠随机分为5组,每组24只。黄连解毒汤高剂量组(0.086 g/mL)、中剂量组(0.043 g/mL)和低剂量组(0.026 g/mL)、空白组(蒸馏水)和抗生素组(0.134 g/mL),分别用相应药物进行灌胃,其间临床观察腹泻情况,服药14 d后分别采集粪便样本。运用GC-MS方法对粪便样本中乳酸、异丁酸、异戊酸、戊酸、萘的含量测定,并进行统计分析。结果抗生素组和黄连解毒汤高剂量组均出现明显腹泻,中剂量、低剂量组和空白组实验动物未出现腹泻。与空白组比较,黄连解毒汤高剂量组和抗生素组粪便中乳酸、异丁酸、异戊酸、戊酸、萘含量差异有统计学意义(P<0.05),黄连解毒汤中剂量组样本异戊酸、戊酸、萘差异有统计学意义(P<0.05);与低剂量组比较,黄连解毒汤高剂量组和抗生素组实验动物中乳酸、异丁酸、异戊酸、戊酸和萘含量差异有统计学意义(P<0.05),黄连解毒汤中剂量组异戊酸和戊酸含量差异有统计学意义(P<0.05)。结论黄连解毒汤和抗生素对实验动物脂肪酸代谢有一定的影响,研究结果可为黄连解毒汤临床合理应用提供实验依据。
Objective To investigate the effects of different doses of Huanglianjiedu decoction(HLJD)on fatty acid metabolism in SD rats,and to provide some reference materials on the clinical rational use of HLJD.Methods Healthy SD rats were randomly divided into HLJD high-dose group(0.086 g/mL,n=24),medium-dose group(0.043 g/mL,n=24)and low-dose group(0.026 g/mL,n=24),the distilled water group(distilled water,n=24)and the antibiotic group(0.134 g/mL,n=24).Corresponding drugs were given with gastric administration in each group.Diarrhea was observed.After 14 d,the feces samples were collected.The contents of lactic acid,isobutyric acid,isovaleric acid,valeric acid and naphthalene in the samples were determined by GC-MS.Experimental data was statistically analyzed.Results There were obvious diarrhea in the antibiotic group and the HLJD high-dose group,while there was no diarrhea in the HLJD medium-dose group,the HLJD low-dose group and the blank control group.Compared with the blank group,there were significant reductions in the contents of lactic acid,isobutyric acid,isovaleric acid,valeric acid and naphthalene from feces of high-dose group and antibiotic group(P<0.05);and significant reductions in the contents of isovaleric acid,valeric acid and naphthalene in medium-dose group(P<0.05).The contents of actic acid,isobutyric acid,isovaleric acid,valeric acid and naphthalene in the high-dose group and antibiotic group were significantly reduced compared with the low-dose group(P<0.05).The contents of isovaleric acid and valeric acid in the medium-dose group were significantly lower than the low-dose group(P<0.05).Conclusion HLJD has a certain effect on host fatty acid metabolism,providing experimental basis for clinical application in HLJD.
