摘要
目的:测定黔产钩藤不同部位中总黄酮的含量。方法:以芦丁为对照品,采用紫外分光光度法在510 nm下,对钩藤的皮、叶、钩、茎、木质部、薄壁组织中总黄酮进行含量测定。结果:芦丁对照品在0.3428μg/mL~2.0565μg/mL范围内线性关系良好,Y=0.3896X+0.03(r=0.9994);加样回收率96.34%,RSD=2.17(n=6);钩藤不同部位中皮的含量最高,为196.26μg/g。薄壁组织含量最低,为38.70μg/g。如下:皮>叶>钩>茎>木质部>薄壁组织。结论:建立的紫外分光光度法操作简便,重复性良好,可用于钩藤中总黄酮的含量测定;造成黄酮类化合物在钩藤不同部位分布存在差异的原因,可能与黄酮类的生物合成途径有关,植物的生物合成途径需要相应的酶发生作用,该结果可以为钩藤的更进一步研究与开发利用提供科学依据。
In this study we determined the content of total flavonoids in different parts of Uncaria rhynchophylla infrom Guizhou.The content of total flavonoids in the bark,leaves,hooks,stems,xylem and parenchyma of U.rhynchophylla was determined at 510 nm by ultraviolet spectrophotometry with rutin as the standard control.The results showed that rutin had a good linearity in the range of 0.3428 μg/mL to 2.0565 μg/mL, Y=0.3896X+0.03(r=0.9994);the recovery rate was 96.34%,RSD was 2.17( n =6).The content of total flavonoids in different parts of U.rhynchophylla was different,and its content in the bark was the highest (196.26 μg/g),while that in the parenchyma was the lowest (38.70 μg/g).The content of total flavonoids ranked in the following order:bark>leaves>hooks>stems>xylem>parenchyma.This method is easy to operate and has good reproducibility,and can be used to determine the content of total flavonoids in U.rhynchophylla .The difference in the distribution of flavonoids in different parts of U.rhynchophylla could be related to the biosynthesis pathway of flavonoids,for the biosynthesis pathway of plants requires corresponding enzymes.This studyhas provided scientific basis for the development and utilization of U.rhynchophylla.
