摘要
为确定禽巴氏杆菌活菌数较高范围的致病力及白头翁汤对禽巴氏杆菌的抗感染作用,综合应用分光光度法和平板计数法绘出禽巴氏杆菌生长曲线,确定禽巴氏杆菌活菌数较高的菌液OD600nm值范围。序贯法测定禽巴氏杆菌半数致死量(LD_(50)),急性毒试验检测白头翁汤中药方剂安全性,体外药敏试验检测白头翁汤对禽巴氏杆菌的抑菌作用。研究发现菌液OD600nm值与活菌数存在的相关性不高,仅在稳定期以前存在增长趋势的正相关。试验筛选出禽巴氏杆菌活菌数较高的OD600nm值范围是0.30~0.55。序贯法测出禽巴氏杆菌的LD_(50)值为5.0×10~7CFU/mL。急性毒试验发现白头翁汤中药方剂LD_(50)大于5 000 mg/kg,说明该中药方剂无毒。白头翁汤对禽巴氏杆菌体外最小抑菌浓度为3.91 mg/mL,表明禽巴氏杆菌对白头翁汤中药方剂高度敏感。
To determine the pathogenicity of Pasteurella multocida and the anti infection ability of Pulsatilla Decoction to Pasteurella multocida,the growth curve of Pasteurella multocida was plotted by spectrophotometric method and plate counting method.The range of OD 600nm value of the bacteria with high living Pasteurella was determined.The sequential method was used to measure the half lethal dose of Pasteurella LD 50,and the acute toxicity test was used to detect the safety of the Chinese medicine prescription.In vitro,the drug sensitivity test was used to detect the antiinfection of Pasteurella.It is found that the correlation between OD 600nm value and viable count is not high,and there is a positive correlation in the growth trend only before stable period.In this experiment,the range of OD 600nm with a higher number of viable strains was 0.3-0.55.The median lethal dose of Pasteurella multocida measured by sequential method was 5.0×10^7CFU/mL.The acute toxicity test showed that the LD 50 of Pulsatilla Decoction is more than 5 000 mg/kg,and the Chinese medicine prescription is non-toxic.The minimum inhibitory concentration of Pulsatilla Decoction against Pasteurella multocida is 3.91 mg/mL,indicating that Pasteurella multocida is highly sensitive to Chinese herbal medicine of Pulsatilla Decoction.
