川芎嗪对SD大鼠心室肌细胞钠离子通道电流的影响 被引量:2

Effects of tetramethylpyrazine on sodium channel currents in ventricular myocytes of SD rats

摘要 采取主动脉逆向灌流酶解法获得SD大鼠单个心室肌细胞,运用全细胞膜片钳技术记录钠电流(7Na),以细胞外液灌流给药,分别记录不同浓度(0-400μmol·L^-1)川芎嗪作用前后INa的变化。结果表明:50和200μmol·L^-1的川芎嗪使的4V曲线显著上移,使峰值钠电流极显著下降(P<0.01,n=6),但I-V曲线的轨迹趋势未发生变化;川芎嗪使的激活曲线右移、失活曲线左移、失活后恢复曲线左移,半数激活电压(Vi/2,ac)给药前与给药后差异显著(P<0.05,n=6),半数失活电压(V1/2,in)和恢复时间t给药前与给药后差异显著(P<0.05,n=6)。这一研究提示川芎嗪浓度依赖性阻滞大鼠心室肌细胞INa,但对通道的动力学特征无显著影响。 Single ventricular myocytes of SD rats were obtained by reverse perfusion of aorta. The sodium current (INa)was recorded by whole-cell patch-clamp technique. The changes of ga were recorded before and after treatment with different concentrations (0-400μmol·L^-1) of tetramethylpyrazine. The results showed that: Under the influence of 50 and 200μmol·L^-1tetramethylpyrazine, the peak sodium current decreased significantly and the I-V curve of INa increased significantly (P<0.01, n=6). While the track trend of I-V curve did not change. The steady-state activation curve shifted to the right, the steady-state inactivation curve shifted to the left and the recovery curve shifted to the left under the influence of tetramethylpyrazine. The difference of half activation voltage (V1/2,ac), half inactivation voltage (V1/2,in)and recovery time (z) before and after administration was significant (P<0.05, n=6). That was to say, tetramethylpyrazine blocked INa of rat ventricular myocytes in a concentration-dependent manner, but had no significant effect on the kinetic characteristics of the channel.
