胶体金免疫层析法快速测定蜂胶及蜂胶乙醇提取物中氯霉素残留 被引量:6

摘要 建立了胶体金免疫层析法快速测定蜂胶及蜂胶乙醇提取物中氯霉素残留的检测方法。样品在碱性条件下溶解稀释,20%高氯酸溶液除杂过滤后,加入碱化剂适量,经乙酸乙酯提取,盐析净化后,再用乙酸乙酯提取吹干,加入正己烷和复溶液溶解后,用胶体金快速检测试剂板快速检测。结果表明,胶体金免疫层析法对蜂胶及蜂胶提取物中氯霉素残留量的检测限分别为0.1μg/kg和0.2μg/kg。经高效液相色谱-串联质谱确证,假阳性率和假阴性率均为0%。该方法操作简单、灵敏度高、准确可靠,可用于加工蜂胶及蜂胶乙醇提取物过程中对氯霉素残留量的质量控制。 A method for rapid determination of chloramphenicol residues in propolis and ethanol extracts of propolis by colloidal gold immunochromatography assay was established. The sample was dissolved and diluted under alkaline conditions. 20% perchloric acid was used to remove the impurities. After being filtered, proper amount of alkalization agent was added subsequently. The residue was extracted with ethyl acetate. After salting out, the supernatant was extracted with ethyl acetate again. The extract was dried. The residue was redissolved with hexane and complex solution. Then, the solvent extract was analyzed by colloidal gold test strip for rapid detection. The results showed that the limits of detection for chloramphenicol residues in the propolis and ethanol extracts of propolis were 0.1 μg/kg and 0.2μg/kg respectively by colloidal gold immunochromatography assay. It was confirmed by high performance liquid chromatography coupled with tandem mass spectrometry, and both false positive rate and false negative rate was 0%. This method is simple, effective and sensitive, which is suitable for the quality control of chloramphenicol residues during the processing of propolis and ethanol extracts of propolis.
