北五味子中总木脂素类成分的抗氧化及抗炎活性研究 被引量:29

Antioxidant and Anti-inflammatory Activity Study of Total Lignin from Schisandra chinensis(Turcz.)Baill.

摘要 对北五味子中总木脂素类成分进行纯化并研究其抗氧化和抗炎活性。利用大孔吸附树脂纯化五味子总木脂素,采用总抗氧化活性法、超氧阴离子自由基清除率法测定总木脂素类成分的抗氧化活性,应用酶联免疫吸附测试(enzymelinkedimmunosorbentassay,ELISA)和一氧化氮(NO)试剂盒考察五味子总木脂素类成分对脂多糖(lipopolysaccharide,LPS)诱导的小鼠巨噬细胞分泌肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-10(Interleukinin-10,IL-10)、NO的影响。抗氧化活性研究方面,与同等浓度的VC对照品相比,北五味子总木脂素类成分的总抗氧化能力是维生素C(VC)的1/7左右,清除超氧阴离子自由基的强度是VC的1/6左右,说明其具有良好的抗氧化活性。抗炎活性研究方面,北五味子总木脂素类成分可显著促进小鼠巨噬细胞分泌IL-10,抑制TNF-α、NO,说明其具有良好的抗炎活性。北五味子总木脂素类成分具有良好的抗氧化能力和抗炎活性,为进一步研究及开发北五味子提供理论支持。 The purification process of total lignin in Schisandra chinensis (Turcz.)Baill. was optimized and the antioxidant and anti-inflammatory activities was studied. The total lignin of of Schisandra chinensis (Turcz.)Baill. was putified by macroporous resin. Total antioxidant activity method and superoxide anionic radical removal method were used to determine the antioxidant activity of the purified total lignin. The effects of lipopolysaccharide(LPS)-induced mouse macrophages on TNF-α,IL-10 and NO secretion were investigated by using ELISA kit. In terms of antioxidant activity,the total antioxidant capacity of total lignans of Schisandra chinensis (Turcz.) Baill. was about 1/7 of the same concentration of VC reference. The intensity of scavenging superoxide anion radicals was 1/6 of VC reference. So it has good antioxidant activity. In terms of anti-inflammatory activity,the total lignans of Schisandra chinensis (Turcz.) Baill. could significantly promote the secretion of IL-10 by RAW264.7,while inhibit TNF-α and NO,indicating that it has good anti-inflammatory activity. The total lignin in Schisandra chinensis (Turcz.) Baill. had good anti-oxidation ability and anti-inflammatory activity. It will provide theoretical support for further research and development of Schisandra.
