盐藻固体培养基分离纯化方法的建立

摘要 研究建立了一种基于固体培养基分离纯化盐藻藻种的方法,通过对不同接种温度、琼脂浓度、培养基盐度、盐藻接种密度的实验,获得了适合固体培养基分离纯化盐藻的接种温度范围为40℃~45℃,琼脂浓度范围为0. 6%~1. 0%,培养基盐度范围为6%~8%,接种盐藻细胞浓度范围为1×10~3个/L~1×10~4个/L。该方法相比于传统的稀释分离、微细管分离等方法,具有操作简单、易操作、纯化成功率高、不受盐藻细胞游动和高盐度培养基等条件的影响等优点。该研究为盐藻生产提供了一个简单有效的分离纯化盐藻藻种的方法。 A method for microalgae Dunaliella spp. isolation and purification on solid medium was established in this article. Through the experiment on different inoculation temperature, agar concentration, medium salinity and inoculation cell density, the optimum inoculation temperature, agar concentration, medium salinity and inoculation cell density are in a range of 40 ℃~45 ℃, 6%~8% and 1×10 3 cells/L^1×10 4 cells/L respectively. Compared with traditional dilution and microtubule isolation methods, this method is easy and simple to operate, high probability of isolation and purification is obtained, and there is no any influence from swimming cells of Dunaliella spp. and high salinity medium. This study provides a sample and efficient method for isolation and purification of the algae stock in microalgae Dunaliella spp. production.
