摘要 目的:研究人脐带间充质干细胞(umbilical cord mesenchymal stem cells,UMSCs)对恶性神经胶质瘤(malignant glioma,MG)细胞株U87MG细胞簇形成及mdr1基因/P-糖蛋白(P-glycoprotein,P-gp)表达的影响。方法:以U87MG细胞为靶细胞,将UMSCs与U87MG间接共培养,用实时荧光定量逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)检测mdr1基因mRNA的表达,流式细胞仪测定P-gp的表达;细胞簇形成法检测细胞的自我更新和增殖能力。结果:随着UMSCs上清液浓度增加,U87MG细胞簇数目明显减少;mdr1/P-gp表达量明显增高(P<0.05,P<0.01)。结论:UMSCs可诱导U87MG细胞凋亡,使多耐药基因mdr1表达增强,基因代谢产物P-gp阳性率增加。 Objective:To investigate and compare the effects of umbilical cord mesenchymal stell cells(UMSCs) on human U87 malignant glioma(MG) mass cells,mdr1 and P-glycoprotein(P-gp) expression.Methods:The U87 MG cells were used as the target cells.The mdr1 mRNA expression was determined by real-time reverse transcription-polymerase chain reaction(RT-PCR),and the expression of P-gp was detected by flow cytometry.The self-renewal and proliferating potential of the cells were examined with colony-forming units assay.Results:Different concentrations of UMSCs significantly induced U87 MG mass cells to apoptosis.The mdr1 and P-gp expression obviously increased(P<0.05,P<0.01).Conclusion:UMSCs is able to induce the apoptosis of U87 MG cells,and increaes the expression of mdr1/P-gp.
