摘要
目的:以莲藕渣为实验材料,分别用酸法、碱法和水法提取获得多糖,研究不同提取方法对莲藕渣多糖提取率、多糖含量、糖醛酸含量和蛋白质含量的影响。方法:采用苯酚-硫酸法测定多糖含量,间羟基苯酚法测定糖醛酸含量。结果表明:采用酸提法多糖得率最高,为10.9%,多糖含量为87.97±1.71%,但是酸提法会导致糖苷键断裂,需严格控制酸度,难度较大;碱提法效率最低。综合比较,水提法较为简单,效率、生物活性高,提取莲藕渣多糖最为合适。
Objective:To study the effects of different extraction methods on the extraction rate,polysaccharide content,uronic acid content and protein content of lotus root residues by using acid method,alkali method and water method.Methods:The content of polysaccharide was determined by phenol-sulfuric acid method,and the content of uronic acid was determined by m-hydroxy phenol method.Results:The yield of polysaccharide extracted by acid methods was the highest, with 10.9% and the content of polysaccharide was 87.97 ± 1.71 %.However,the acid leaching could lead to the cleavage of the glycosidic bond.It was difficult to control of acidity.Comprehensive comparison,water extraction is relatively simple,high efficiency,high biological activity,and therefore it was the most appropriate to extraction lotus root polysaccharides.
