摘要
目的:探讨黄连解毒汤对KM小鼠T细胞组成及肠粘膜中细胞因子表达的影响。方法:用黄连解毒汤灌胃小鼠7d和14d后分别采集相关样本,ELISA法测定小鼠回盲段肠粘液中sIgA、IL-6及IL-22表达水平;流式细胞仪检测小鼠脾脏中CD3^+T、CD4^+T和CD8^+T含量。结果:与空白组相比,黄连解毒汤给药组CD3^+T含量明显升高,CD4^+T和CD8^+T含量则明显降低,灌胃7d与14d对于给药组CD3^+T、CD4^+T和CD8^+T含量无明显变化;黄连解毒汤给药组sIgA浓度明显升高,IL-6含量则明显降低,灌胃7d与14d对于给药组sIgA含量无明显变化,IL-6变化不明显,IL-22于7d升高而于14d降低,具有极显著差异(P<0.01)。结论:黄连解毒汤能促进CD3^+T和sIgA分泌,抑制CD4^+T、CD8^+T和IL-6分泌和表达,对IL-22的表达则具有先促进后抑制的作用。
Objective: To investigate the effect of Huanglian Jiedu Decoction on T cell composition and cytokine expression in intestinal mucosa of KM mice. Methods: The expression levels of sIgA,IL-6 and IL-22 in ileocecal intestinal mucus of mice were detected by Elisa with Huanglian Jiedu Decoction for 7 days and 14 days, respectively. Flow cytometry assay was used to detect the levels of CD3^+T CD4^+T and CD8^+T in mouse spleen. Results: Compared with the blank group, the content of CD3^+T in the Huanglian Jiedu Decoction group was obviously increased and the content of CD4^+T and CD8^+T decreased significantly. The content of CD3^+T, CD4^+T and CD8^+T in the administration group was not obviously changed after intragastric administration for 7 d and 14 d. The concentration of sIgA of the Huanglian Jiedu Decoction group increased obviously, the content of IL-6 was observed decreased. After 7 d and 14 d, the content of sIgA and IL-6 showed no significant change. The concentration of IL-22 increased after 7 d and decreased significantly after 14 d(P< 0.01). Conclusion: Huanglian Jiedu Decoction can promote the secretion of CD3^+T and sIgA, inhibit the secretion and expression of CD4^+T, CD8^+T and IL-6. For IL-22 expression,it was promoted first and inhibited later.
