摘要
以新疆阿克苏地区采集的野生黄伞菌(Pholiota adiposa)为研究对象,采用形态学和分子生物学相结合方法确定其种类,并对其生长适宜的碳源、氮源、无机盐、光照、温度、pH值进行筛选。结果表明,野外采集的子实体标本经分离纯化获得其纯培养物,通过ITS区段序列分析,结合形态学特征,确定该野生菌为黄伞菌;黄伞菌丝在加富培养基上的生长势优于在PDA培养基上的生长势;黄伞菌丝生长的适宜碳源为葡萄糖,适宜氮源为蛋白胨;添加MgSO4时对菌丝生长的影响较明显;无光条件下菌丝生长较好;菌丝生长的最适温度为25℃;最适pH值为8.0。
Pholiota adiposa collected in Aksu area of Xinjiang was used as the search object,and its species was identified by the combination method of morphological and molecular biology,and suitable carbon source,nitrogen source,inorganic salt,light,temperature and pH were screened.The experimental results showed that the wild strain was isolated and purified to obtain pure culture.According to the ITS sequence analysis and by combining the morphological characteristics,the wild fungus was determined to be a yellow umbrella (Pholiota adiposa). The growth potential of hyphae in enriched medium was better than that in PDA medium.The suitable carbon source for hyphae growth was glucose and the suitable nitrogen source was peptone.The added MgSO 4 had an obvious effect on mycelial growth;Mycelium growth was better under no light condition;The optimal temperature for mycelium was 25 ℃ and the optimal pH for mycelium growth was 8.0.
