摘要
从高粱转录组数据筛选获得SbDCLs基因全长序列,并结合生物信息学方法在高粱的全基因组中鉴定出了6个SbDCLs基因,分布于SBI-01,SBI-03和SBI-06上。其中,SbDCL2a和SbDCL2b在SBI-01上串联重复排列,比较二者基因结构和蛋白质结构域推测SbDCL2a很可能是由SbDCL2b小范围复制所致。SbDCLs蛋白质均为两性蛋白且没有信号肽,亚细胞定位均在细胞核上,二级结构以α-螺旋和无规则卷曲为主。通过对高粱、水稻、玉米、拟南芥、大豆、小麦、藜麦、甜菜、谷子的DCL蛋白质序列进行系统进化分析,结果表明,DCL存在4个分枝:DCL1,DCL2,DCL3和DCL4;DCL在进化上出现了单双子叶的分化;DCL蛋白编码基因进化具有纯化效应。对SbDCLs基因进行表达分析,结果显示,SbDCLs在高粱穗部、茎节中部、花梗和旗叶茎节表达都比较强烈;SbDCL2a,SbDCL1和SbDCL3a是高度表达基因,而且在生殖器官或组织中相对于营养器官具有更高的表达水平,说明SbDCLs基因对高粱营养生长和生殖发育具有调控作用。SbDCL2b在高粱的整个生育期表达水平均较低,但是在胚珠和花粉中拥有较高的表达水平,可能参与生殖发育调控。
The full-length sequence of SbDCLs was obtained from sorghum transcriptome data,and six SbDCLs genes were identified in Sorghum bicolor genome by bioinformatics methods,which were distributed on SBI-01,SBI-03 and SBI-06.Among them,SbDCL2a and SbDCL2b were repeatedly arranged in tandem on SBI-01.Comparing the two gene structures and protein domains,it was speculated that SbDCL2a was likely to be caused by small-scale replication of SbDCL2b.The SbDCLs proteins were amphiphilic proteins and had no signal peptide.The subcellular localization was on the nucleus,and the secondary structure was dominated by α-helix and random coil.Phylogenetic analysis of DCL protein sequences of sorghum,rice,maize,Arabidopsis,soybean,wheat,quinoa,sugar beet and millet showed that DCL had four branches: DCL1,DCL2,DCL3 and DCL4;DCL had monocotyledon and dicotyledon evolution;DCL protein coding gene evolution had purification effect.SbDCLs gene expression analysis showed that SbDCLs were strongly expressed in ear,middle stem node,pedicel and flag leaf stem node of sorghum.SbDCL2a,SbDCL1 and SbDCL3a were highly expressed genes and had higher expression level in reproductive organs or tissues than in vegetative organs,indicating that SbDCLs gene had a regulatory effect on vegetative growth and reproductive development of sorghum.The expression level of SbDCL2b is lower in the whole growth stage of sorghum,but it has higher expression level in ovule and pollen,which may be involved in the regulation of reproductive development.
