摘要
目的:观察人参对异丙肾上腺素(ISO)诱导的大鼠心肌缺血的保护作用。方法:50只SD大鼠随机分成5组:正常对照组、模型组、人参低(以生药材计,405 mg·kg^-1·d^-1)、高(以生药材计,1 618 mg·kg^-1·d^-1,H-G)剂量组、维拉帕米(2 mg·kg^-1·d^-1)组,每组10只。人参各剂量组灌胃给药,维拉帕米组腹腔注射,连续7 d,正常对照组和模型组给予等量蒸馏水,除正常对照组外,其余各组大鼠皮下注射ISO(85 mg·kg^-1·d^-1)建立急性心肌缺血模型,连续2 d。观察大鼠心电图心率和J点的变化,测定血浆乳酸脱氢酶(LDH)和肌酸激酶(CK)活性水平及心肌组织超氧化物歧化酶(SOD)、丙二醛(MDA)的含量,观察大鼠心肌组织病理形态学的改变。结果:人参能显著降低ISO所致心肌缺血大鼠的心率、J点的抬高、血浆中LDH和CK活性水平及心肌组织MDA含量,升高SOD的活性水平(P<0.05或P<0.01),同时明显减轻缺血心肌的病理损伤程度。人参各剂量的CK、高剂量组的LDH、SOD及低剂量组的MDA与Ver组比较差异有统计学意义(P<0.05或P<0.01)。结论:人参对ISO所致的大鼠急性心肌缺血有明显的保护作用,其机制可能与抗氧化、减少氧自由基生成、减少心肌酶的漏出有关。
Objective:To observe the protective effect of Panax ginseng C.A.Mey on isoproterenol(ISO)-induced myocardial ischemia in rats.Methods:Fifty SD rats were randomly divided into five groups:the normal control group,the model group,Panax ginseng C.A.Mey low(calculating with crude medicinal materials,405 mg·kg^-1·d^-1)and high(calculating with crude medicinal materials,1 618 mg·kg^-1·d^-1)dose groups and verapamil group(2 mg·kg^-1·d^-1)with 10 rats in each.Panax ginseng C.A.Mey was given orally,verapamil was injected intraperitoneally,and the normal control group and the model group were given equal amount of distilled water for 7 days.Except for the normal control group,the other groups were subcutaneously injected with ISO(85 mg·kg^-1·d^-1)for 2 days to establish acute myocardial ischemia model.The changes of heart rate and J-point of ECG in rats were observed,the activities of lactate dehydrogenase(LDH)and creatine kinase(CK)in plasma and the contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in myocardium were measured,and the pathomorphological changes of myocardium were observed.Results:Panax ginseng C.A.Mey could significantly decrease heart rate,J point and the levels of CK,LDH and MDA,increase the activity of SOD(P<0.05 or P<0.01),and reduce myocardial cell morphology changes in ISO-induced myocardial ischemia rats.The CK in different dose groups of Panax ginseng C.A.Mey,the LDH and SOD in high group and the MDA in low group had significant differences from those in Ver group(P<0.05 or P<0.01).Conclusion:Panax ginseng C.A.Mey has obvious protective effect on acute myocardial ischemia induced by ISO in rats,which may be associated with inhibiting oxidative stress,decreasing oxygen free radical production and reducing leakage of myocardial enzymes.
