摘要
为建立滇柴胡中槲皮素和异鼠李素含量测定方法.采用高效液相色谱法测定其含量,检测波长360nm,流动相为V(甲醇)∶V(0.4%磷酸)=45∶55,流速1.0mL/min.结果表明:槲皮素在5.18~207.20μg/mL(r=0.9998)有良好线性关系,检出限0.1260μg/mL,定量限0.4201μg/mL,加标回收率92.4%,RSD为0.51%;异鼠李素在1.026~20.520μg/mL(r=0.9997)有良好线性关系,检出限0.1654μg/mL,定量限0.5516μg/mL,加标回收率94.2%,RSD为1.38%.由此说明,HPLC法能快速、准确地测定滇柴胡中槲皮素和异鼠李素的含量.
To establish a method for determining the content of quercetin and isorhamnetin in the Dianchaihu. HPLC was used with the detection wavelength 360 nm,the flow phase V ( methanol )∶ V (0.4% phosphoric acid )= 45∶ 55 and the flow speed 1.0 mL/min.The result showed that Quercetin in 5.18-207.20 μg/mL ( r =0.999 8) has a good linear relationship within the scope of detection limit of 0.126 0 μg/mL,quantitative limit of 0.420 1 μg/mL and the average recovery rate of quercetin 92.4%. Isorhamnetin at 1.026-20.520 μg/mL( r =0.999 7) has a good linear relationship within the scope of a detection limit of 0.165 4 μg/mL, quantitative limit of 0.551 6 μg/mL and the average recovery rate of isorhamnetin 94.2% with RSD of 0.51% and 1.38%( n =6) respectively. HPLC method can quickly and accurately determine the content of quercetin and isorhamnetin in the Dianchaihu.
