正交实验在蓝莓组培培养基筛选过程中的应用 被引量:5

摘要 诱导蓝莓丛生芽,筛选出适合蓝莓丛生芽诱导、继代增殖和试管苗生根的最佳培养基。设计正交试验,利用蓝莓幼嫩生长新梢为材料,对蓝莓的丛生芽诱导、增殖和试管苗生根技术进行了研究。诱导蓝莓丛生芽萌发的最适宜培养基为1/2WPM+0.5 mg/L IBA+2.0 mg/L ZT+pH 5.0,诱导丛生芽发芽率为85%,并且长势良好,存活率也比较高;蓝莓丛生芽增殖的适宜培养基为1/2MS+1.0 mg/L ZT+1.5 mg/L 6-BA,丛生芽增殖倍数高达150%,分化的苗数比较多,生长比较旺盛,而且比较粗和高;蓝莓试管苗生根的适宜培养基为改良1/2WPM+1.0 mg/L ZT+0.8 mg/L NAA+pH 5.0+0.25%活性炭,试管苗生根率高达86%,根系比较粗状,分支也比较多。 To screen out the optimum media for the clustered shoot inducing,multiplication and rooting of blueberry plantlets by using orthogonal experimental design.The tender shoot of blueberry was adopted as experimental material.Media in different combination and different concentrations based on orthogonal experimental design were applied to optimize the shoot inducing,multiplication and rooting of blueberry plantlets.Results and conclusion:The results shows that the optimal medium for inducing the sprouting of blueberry clustered shoots is 1/2WPM+0.5 mg/L IBA+2.0 mg/L ZT +pH 5.0.Under this condition the germination percentage is 85% followed by better growth and higher survival rate of blueberry.The most suitable medium for the multiplication of blueberry shoots is 1/2MS+1.0 mg/L ZT+ 1.5 mg/L 6-BA,with a proliferation factor of up to 150%,and under this condition,the plantlets are sturdier and higher.The most suitable medium for rooting of in vitro blueberry plantlet is improved 1/2WPM+1.0 mg/L ZT+0.8 mg/L NAA+pH 5.0+0.25% activated carbon which is followed by rooting rate of 86% with more sturdy roots branches.
