摘要
目的观察单味中药白芷、吴茱萸、川芎对偏头痛模型大鼠神经递质及脑干C-FOS表达的影响。方法将48只Wistar大鼠随机分为6组(每组8只):空白组、模型组、白芷治疗组、吴茱萸治疗组、川芎治疗组、西比灵治疗组(药物对照)。各治疗组分别按0.79g/kg、0.40g/kg、0.79g/kg、0.43mg/kg干预,模型及空白组灌胃相同体积生理盐水,连续7d。除空白组外,均于末次灌胃1h后,硝酸甘油10mg/kg皮下注射方法制备大鼠偏头痛模型。4h时采血,ELISA法检测血清脑啡肽(Enkephalin,ENK)、β内啡肽(β-Endorphin,β-EP)含量;免疫组化方法检测脑干组织C-FOS表达;并观察各组大鼠行为学表现。结果与空白组比较,模型组动物血清ENK含量显著增高(P<0.05),血清β-EP含量显著降低(P<0.05);与模型组比较,各治疗组ENK含量及C-FOS表达降低(P<0.05),β-EP含量显著增高(P<0.05);川芎组ENK含量低于白芷组(P<0.05),川芎组C-FOS表达低于吴茱萸组(P<0.05)。结论3种中药均能改善偏头痛动物双耳变红、挠头、爬笼、竖耳、皮肤发绀等行为学表现,减少ENK分泌,增加β-EP含量,降低C-FOS表达。
Objective To observe the effects of single traditional Chinese medicine (TCM)-Radix Angelicae Dahuricae, Fructus Evodiae and Rhizoma Chuanxiong on neurotransmitters and the expression of C-FOS in the brainstem of migraine model rats. Methods A total of 48 Wistar rats were randomly divided into 6 groups (each group of 8 rats): a blank group, a model group, a Radix Angelicae Dahuricae group (BZ group, 0.79 g/kg), a Fructus Evodiae group (WZY group, 0.40 g/kg), a Rhizoma Chuanxiong group (CX group, 0.79 g/kg) and a drug control group (XBL group, 0.43 mg/kg). Each treatment group was given its respective medicine, while the blank group and the model group were given the same volume of normal saline by gavage, for 7 d. Except for the blank group, the rat model of migraine was prepared by subcutaneous injection of nitroglycerin 10 mg/kg at 1 h after the last gavage. At the end of 4 h, blood was collected. The content of enkephalin (ENK) and β-endorphin (β-EP) in serum was tested by ELISA, and the expression of C-FOS in brainstem was detected by immunohistochemistry (IHC). And the behavior of each group of rats was observed. Results Compared with the blank group, the content of ENK in serum in the model group was significantly increased (P<0.05), and the content of β-EP in serum was significantly decreased (P<0.05). Compared with the model group, the content of ENK and the expression of C-FOS in each treatment group were decreased obviously (P<0.05), and the content of β-EP was increased significantly (P<0.05). The ENK content of the CX group was lower than that in the BZ group (P<0.05), and the C-FOS expression of the CX group was lower than that in the WZY group (P<0.05). Conclusion The 3 kinds of TCM can improve the behavioral performance of migraine animals such as red ears, head scratching, climbing cage, erect ears and cyanosis, reduce the secretion of ENK, increase the content of β-EP and decrease the expression of C-FOS.
