林蛙低聚肽的体外抗氧化与ACE抑制作用 被引量:5

摘要 以林蛙为原材料,通过酶解获得林蛙低聚肽,测定了其相对分子质量分布,又从DPPH自由基、羟基自由基、超氧阴离子自由基和还原能力四个方面对其抗氧化作用较全面的进行了评价;另外从林蛙低聚肽对血管紧张素转化酶(angiotensin converting enzyme,ACE)的抑制作用对其降血压活性进行了分析;结果显示:林蛙低聚肽的相对分子质量分布大多为1000Da以下(高达85%),其中140~500Da之间的小肽最多(约占52%)。三种自由基半抑制浓度(IC50)分别为:2.3mg/mL、3mg/mL和15mg/mL;浓度为5mg/mL时还原能力与VC浓度0.03mg/mL相当;另外其ACE抑制活性实验中IC50值为0.5mg/mL,林蛙低聚肽的抗氧化能力和ACE抑制活性与浓度都呈现正相关。证明了林蛙低聚肽具有良好的抗氧化作用和ACE抑制能力,为下一阶段相关功能肽段的筛选、纯化奠定良好的基础,为相关抗氧化和降血压等保健类食品的开发利用提供一定的理论依据。 The frog oligopeptide was obtained by enzymatic hydrolysis of Rana chensinensis,and its relative molecular weight was determined.Its anti-oxidation effect was evaluated on DPPH free radicals,hydroxyl radicals,superoxide anion radicals and reducing ability.In addition,the antihypertensive activity of angiotensin-converting enzyme(ACE)was analyzed from the inhibition of angiotensin-converting enzyme(ACE).The results showed that the molecular weight is mostly below 1000 Da(up to 85%),of which 52%were smaller peptides with molecular weight between 140 and 500 Da.The inhibitory concentrations(IC50)of three free radicals were:2.3 mg/mL,3 mg/mL,and 15 mg/mL,respectively.The reducing ability of 5 mg/mL of oligopeptide was equivalent to 0.03 mg/mL VC.In addition,the ACE inhibitory activity was tested.The IC50 value was 0.5 mg/mL,and the antioxidant capacity and ACE inhibitory activity of Rana oligopeptide were positively correlated with the concentration.It is proved that the frog oligopeptide has good antioxidation and ACE inhibition ability.The research lays a good foundation for the screening and purification of relevant functional peptides in the next stage.In addition,it provides a theoretical basis for the development and utilization of the frog oligopeptide in functional foods such as anti-oxidation and blood pressure lowering.
