摘要
目的建立高效液相色谱-电雾式检测器(HPLC-CAD)法同时测定风湿定片中八角枫苷A、甘草酸、水杨苷、甘草次酸、毒藜碱、丹皮酚、欧前胡素及异欧前胡素的含量,为风湿定片的质量研究提供理论依据。方法采用HPLC-CAD法,色谱柱为Thermo Hypersil GOLD C18(250 mm×4.6mm,5μm),甲醇(A)-20 mmol·L^-1甲酸铵(甲酸调pH至5.0)(B)为流动相,梯度洗脱,流量1.0mL·min-1,柱温30℃,CAD雾化器温度为35℃,进样量20μL。结果八角枫苷A、甘草酸、甘草苷、水杨苷、毒藜碱、丹皮酚、欧前胡素及异欧前胡素的质量浓度分别在0.28~5.60(r=0.9992),0.36~7.20(r=0.9993),0.53~10.60(r=0.9996),0.25~5.00(r=0.9990),1.94~38.80(r=0.9994),2.02~40.40(r=0.9994),0.21~4.20(r=0.9991),0.18~3.60μg·mL^-1(r=0.9991)内线性关系良好。八角枫苷A、甘草酸、水杨苷、甘草次酸、毒藜碱、丹皮酚、欧前胡素及异欧前胡素的平均加样回收率分别为99.3%、98.8%、99.0%、98.5%、99.1%、99.1%、99.0%及98.9%,RSD分别为1.4%、1.3%、0.90%、0.80%、1.1%、1.5%、1.3%及1.2%(n=6)。结论该方法准确性好,灵敏度高,且操作简便,可用于风湿定片中多指标成分的质量控制研究。
Objective To develop an HPLC-CAD method for the simultaneous determination of 8 active ingredients (chinenside A, glycyrrhizic acid, salicin, glycyrrhetinic acid, anabasine, paeonol, imperatorin, and isoimperatorin) in Fengshiding tablets, and provide theoretical basis for its quality study. Methods HPLC-CAD was used. Chromatographic separation was performed on a Thermo Hypersil GOLD C18 (250 mm×4.6 mm, 5 μm) column with methanol (A)-20 mmol·L^-1 ammonium acetate (formic acid tone pH5.0)(B) as the mobile phase for gradient elution. The flow rate was 1.0 mL·min^-1 , the column temperature was maintained at 30℃, the nebulization temperature of the detector was 35℃, and the injection volume was 20 μL. Results Chinenside A, glycyrrhizic acid, salicin, glycyrrhetinic acid, anabasine, paeonol, imperatorin, and isoimperatorin were linearity at 0.28 - 5.60 (r = 0.9992), 0.36 - 7.20 (r = 0.9993), 0.53 - 10.60 (r = 0.9996), 0.25 - 5.00 (r = 0.9990), 1.94 - 38.80 (r = 0.9994), 2.02 - 40.40 (r = 0.9994), 0.21 - 4.20 (r = 0.9991), and 0.18 - 3.60 μg·mL- 1 (r = 0.9991), respectively. The average recoveries were 99.3%, 98.8%, 99.0%, 98.5%, 99.1%, 99.1%, 99.0% and 98.9%, and the RSDs were 1.4%, 1.3%, 0.90%, 0.80%, 1.1%, 1.5%, 1.3% and 1.2%, respectively. Conclusion The method is accurate, sensitive, convenient, and reproducible for the quality control of multicomponents in Fengshiding tablets.
