摘要 目的:联用纤维素酶法和超声提取法提取杠板归中总黄酮,并探究超声辅助酶解法的最佳提取工艺,与其他传统提取方法进行比较,达到工艺优选的目的。方法:通过单因素实验和正交试验优选出超声辅助酶解法提取杠板归中总黄酮的最佳酶解条件。在该条件下利用超声辅助酶解法提取杠板归中总黄酮,提取液用硝酸铝显色法显色,用紫外分光光度计测其吸光度并计算出提取率,结果与其他传统提取方法相比较。结果:超声辅助酶解法提取杠板归中总黄酮的最佳酶解条件为酶浓度0.2mg·mL^-1,酶解时间90min,酶解温度50℃,酶解pH5.0。结论:超声辅助酶解法提取得率高,方法简易、可行,可用于提取杠板归中总黄酮。 Objective: Enzymatic method of cellulose and ultrasonic extraction method are combined to extract the total flavonoids from the Ploygonum Perfoliatum L.. The best technology of ultrasonic assisted enzymatic hydrolysis was explored and compared with other traditional extraction methods in order to achieve the goal of process optimization. Methods: Through the single factor experiment and an orthonormal experiment,the optimal enzymatic hydrolysis conditions of extracting total flavonoids in the Ploygonum Perfoliatum L. were obtained. In this condition,the total flavonoids were extracted by ultrasonically assisted enzymatic extraction and the extracted liquid was colored with aluminum nitrate chromogenic method. Using ultraviolet spectrophotometer to measure the absorbance,then calculate the extraction yield and compared it with other traditional extraction methods. Results: The optimal enzymatic condition of ultrasonic assisted enzymatic hydrolysis as follows: the enzyme concentration was 0. 2 mg·m L-1,the enzymolysis time was 90 min,the enzymolysis temperature was 50 ℃,and the enzymolysis p H was 5. 0. Conclusions: The extraction rate of ultrasonic assisted enzyme was high,and the method was simple and feasible,it can be used to extract the total flavonoids from Ploygonum Perfoliatum L..
