摘要
目的探究茶花粉水提物对酒精性肝损伤大鼠模型肝脏组织中胶原蛋白Ⅰ(COL1A1)、α-平滑肌肌动蛋白(α-SMA)表达水平的影响。方法选取45只雄性SD大鼠,体重200±30g,将大鼠制成酒精性肝损伤模型,并分为:模型组、低剂量茶花粉水提物组和高剂量茶花粉水提物组,每组各15只,另选15只作为对照组。测量大鼠肝脏质量并计算肝指数;检测大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性、肝组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;HE染色观察肝脏组织的病理学改变;使用蛋白质印记检测肝脏组织中COL1A1和α-SMA表达水平。结果与对照组相比,模型组大鼠肝指数、血清ALT,AST水平、肝组织MAD水平,COL1A1和α-SMA蛋白表达水平均显著升高(P<0.05),肝脏组织中SOD水平显著降低(P<0.05);相比模型组,低剂量茶花粉水提物组和高剂量茶花粉水提物组大鼠肝指数、血清ALT,AST水平、肝组织MAD水平,COL1A1和α-SMA蛋白表达水平均显著降低(P<0.05),且高剂量茶花粉水提物组上述指标显著低于低剂量茶花粉水提物组,肝脏组织中SOD水平显著升高,且高剂量茶花粉水提物组显著高于低剂量茶花粉水提物组(P<0.05)。结论茶花粉水提物可以降低酒精性肝损伤大鼠肝脏组织中胶原蛋白Ⅰ(COL1A1)、α-平滑肌肌动蛋白(α-SMA)表达,有效缓解酒精性肝损伤。
Objective To investigate effects of matter distill from tea pollen on expression levels of collagen Ⅰ (COL1A1) and α-smooth muscle actin (α-SMA) in liver tissue of rat models with alcoholic liver injury. Methods 45 male SD rats weighing200±30gwere selected. The rats were made into alcoholic liver injury models. And they were divided into model group, low-dose matter distill from tea pollen group and high-dose matter distill from tea pollengroup,15 for each group.Another15 rats were selected as control group. Rat liver mass was measured.And liver indexeswere calculated. Serum glutamic pyruvic transaminase (ALT) and glutamic oxaloacetic transaminase (AST) activity, superoxide dismutase (SOD) activity of liver tissues and malondialdehyde (MDA) contentsin rats were detected. HE staining was performed to observepathological changes in liver tissues.The expression levels of COL1A1 and α-SMA in liver tissues were detected by Western blot. Results Compared with those in the control group, the liver indexes, serum ALT and AST levels, MAD level of liver tissues, expression levels of COL1A1 and α-SMA proteins in the model group were significantly increased ( P <0.05), while SOD level in liver tissues were significantly decreased ( P <0.05). Compared with those in the model group, the liver indexes, serum ALT and AST levels, MAD level of liver tissues, expression levels of COL1A1 and α-SMA proteins in low-dose matter distill from tea pollen group and high-dose matter distill from tea pollen group were significantly decreased.The above indexes of high-dosematter distill from tea pollengroup were significantly lower than those of low- dosematter distill from tea pollen group. The SOD level in liver tissues was significantly increased. And this of high- dose matter distill from tea pollen group was significantly higher than that of low dosematter distill from tea pollen group ( P <0.05). Conclusion The matter distill from tea pollen can reduce expression of COL1A1andα-SMA in liver tissues of rats with alcoholic liver injury, and effectively alleviate alcoholic liver injury.
