农药残留速测试剂盒在实际应用中的问题分析 被引量:3

摘要 目的了解农药残留速测试剂盒在实际应用中的效果,为农产品质量安全检测提供技术支撑。方法用紫外分光光度法扫描不同厂家试剂盒酶的活性;测试了不同品牌试剂盒对几种农药的抑制率;并对影响抑制率酶的用量、底物浓度、温度等几种因子进行了分析。结果不同品牌试剂盒一级反应时间差异大,对酶的抑制率影响最大;酶的用量在0.10 ml^0.15 ml时,能获得稳定的抑制率;合适的底物浓度能加快酶促反应速度;几种农药在25℃和37℃对酶的抑制率影响差异不显著。结论建议相关部门在大批量使用前应当对选用的农药残留速测试剂盒进行质量评价,尽可能选用同一品牌试剂盒,通过实验制定相应的抑制率标准,以保证检测结果的一致性和可靠性。 Objective To evaluate the effect of rapid detection kit of pesticide residues in practical application,so as to provide technical support for the quality and safety detection of agricultural products.Methods The activity of enzyme from different factories were canned by UV spectrophotometry.The inhibition rate of the test kits from different brands was tested by working on several kinds of agricultural chemicals.The content,substrate concentration,temperature and other factors which will affect the inhibition rate were analyzed.Results The first class reaction of test kits from different brands varied greatly,and the inhibitory rate of enzyme has been affected the most.When the dosage of enzyme was within 0.10 ml-0.15 ml,it got very stable inhibition rate.Proper substrate concentration can accelerate the enzymatic reaction.The differences in inhibition rate of several agricultural chemicals on enzyme were not obviously at 25 ℃ and 37 ℃.Conclusion In order to ensure the reliability of the test results,it is suggested that the relative departments should evaluate the quality of the testing agent kits before they come into use.The test kits should be selected from the same brand as much as possible.The inhibition rate standard should be formulated through experiments to ensure the consistency and reliability of test results.
