摘要
目的:建立同时测定党参固本丸中党参炔苷、苍术内酯Ⅲ、梓醇、毛蕊花糖苷、甲基麦冬烷酮A及甲基麦冬烷酮B 6种活性成分含量的测定方法,考察不同剂量60Co-γ射线辐照前后有效成分含量的变化。方法:采用Agilent Zorbax-C18色谱柱(250 mm×4.6 mm,5μm),以流动相为乙腈-0.4%磷酸溶液进行梯度洗脱,检测波长分别为210,330,220,296 nm,流速为1.0 ml·min-1,柱温为30℃,进样量为10μl。分别选择辐照剂量为2,5,5 k Gy对党参固本丸进行辐照,比较辐照前后党参固本丸中6种活性有效成分含量的变化,并经组间方差分析考察其显著情况。结果:党参炔苷、苍术内酯Ⅲ、梓醇、毛蕊花糖苷、甲基麦冬烷酮A及甲基麦冬烷酮B分别在0.462~18.464μg、1.006~40.250μg、1.794~71.754μg、0.981~39.236μg、0.477~19.072μg及0.450~18.016μg范围内有良好线性(r≥0.999 5)。经2,5,8 k Gy辐照后,党参固本丸所含的有效成分党参炔苷、苍术内酸Ⅲ、梓醇、毛蕊花糖苷、甲基麦冬烷酮A及甲基麦冬烷酮B均有变化,在超过5 k Gy后,苍术内酯Ⅲ辐照前后含量变化显著(P<0.05)。结论:所建立的党参固本丸活性成分测定方法回收率高,重复性好,简单实用,可作为党参固本丸的质控方法。在辐照剂量不超过5 k Gy时,各成分变化不显著(P>0.05),可为党参固本丸辐照灭菌手段提供参考。
Objective: To establish an HPLC method for the simultaneous determination of 6 active ingredients( lobetyolin,atractylenolide Ⅲ,catalpol,verbascoside,methylmethadone A and methylmethadone B) in Dangshen Guben pills,and study the effect of 60Co-γ ray irradiation on the effective components of Dangshen Guben pills. Methods: An Agilent Zorbax-C18 column( 250 mm ×4.6 mm,5 μm) was adopted and the wavelength of UV detection was 210,330,220,296 nm at the flow rate of 1.0 ml·min-1. The mobile phase consisted of acetonitrile-0.4% phosphoric acid with a gradient elution,column temperature was 30℃ and the injection volume was 10 μl.Results: The linear range of lobetyolin,atractylenolide Ⅲ,catalpol,verbascoside,methylmethadone A and methylmethadone B was 0.462-18.464 μg,1.006-40.250 μg,1.794-71.754 μg,0.981-39.236 μg,0.477-19.072 μg and 0.450-18.016 μg( r≥0.9995),respectively. Conclusion: The established method has a high recovery rate,good repeatability,and simple and practical operation,which can be used for the quality control of Dangshen Guben pills. When the radiation dose is not more than 5 kGy,the content change of each component is not significant( P> 0.05),which can provide reference for the sterilization of Dangshen Guben pills.
