摘要
目的优选并确立假蒟中生物碱的最佳提取工艺。方法采用酸性染色比色法测定总生物碱的含量。以总生物碱及浸膏量为指标,采用正交试验法对提取过程中的乙醇用量、乙醇浓度、提取时间及提取次数4个因素进行考察。结果假蒟的最佳提取工艺为10倍量的80%乙醇,回流提取3次,每次1h。结论优选的提取工艺合理、可行,生物碱提取率较高,为假蒟的进一步开发提供理论和实验依据。
Objective To optimize and establish the best extraction process of total alkaloids from Piper sarmentosum Roxb. Methods Total alkaloids of Piper sarmentosum Roxb. was determined by acid dye colorimetry. With the extraction rate of total alkaloids and dried extract amount as the indexes, the orthogonal experiment selected four factors to find the optimum extraction process, including the dosage and concentration of alcohol, the extraction time and the times of extraction. Results The optimum extraction process of Piper sarmentosum Roxb. was extracted by 10 times the amount of 80% alcohol for 3 times,1 hour at a time. Conclusion The optimum extraction process is reasonable and feasible,and a high extraction rate of alkaloid is achieved, which provides theoretical and experimental basis for further pharmaceutical development of Piper sarmentosum Roxb.
