微血管密度、VEGF及受体FLK-1在宫颈癌中的作用及意义 被引量:5

摘要 目的采用Max Vision法免疫组化检测宫颈癌组织中微血管密度(microvessel density,MVD),检测血管内皮生长因子(vascular endothelial growth factor,VEGF)及其受体FLK- 1在宫颈癌中的阳性表达,探讨MVD、VEGF及FLK-1与宫颈癌增殖、发展的关系。方法选取40例宫颈鳞癌(Ib2~IIb期)患者肿瘤组织为研究对象,以癌旁宫颈组织作为对照组(26例)。采用Max Vision法对宫颈癌及癌旁宫颈组织的石蜡切片进行免疫组化染色,对相关组织的MVD、VEGF及FLK-1阳性表达率进行检测,按Orre和Volm等的判断标准进行评价,并对结果进行统计学分析。结果宫颈鳞癌组织的微血管密度(MVD)、VEGF及FLK-1阳性表达率明显高于癌旁宫颈组织,两者间差异有统计学意义;宫颈鳞癌与对照组间MVD、VEGF及FLK-1的表达和肿瘤的临床分期和病理类型相关;VEGF与受体FLK- 1蛋白表达强度差异无统计学意义(P>0.05)。结论 MVD与宫颈癌的增殖与发展密切相关,VEGF及其受体FLK- 1的高表达在宫颈癌的增殖发展中起着重要的作用。 Objective In order to provide theoretical accordance with the treatment of cervical carcinoma,we adopted Max Vision immunohistochemistry to detect the positive expression of microvessel density(MVD),vascular endothelial growth factor(VEGF)and FLK-1 in cervical carcinoma to evaluate the relationships between MVD,VEGF/FLK-1 and the growth and development of cervical carcinoma.Methods The tumor tissues confirmed by pathology from 40 cases of cervical squamous carcinoma patients(Ib2-IIb Phase)were collected as the research object group,and the cervical carcinoma adjacent tissues were collected as the control group(26 cases).We then used MaxVision immunohistochemistry to observe the micro vessel density,and the positive expressions of VEGF and FIK-1.We counted the microvessel density in microscopy using the methods of Orre and Volm.Results The MVD and the ratio of positive expression of VEGF/FLK-1 in cervical carcinoma were higher than the control group,both the two groups had significant difference.MVD/VEGF/FLK-1 of cervical carcinoma had a close relationship with clinical stages and pathological classification.The expressions of VEGF and FLK-1 had no significant difference( P >0.05).Conclusion MVD is closely related to the growth and development of cervical cancer,while VEGF and its receptor FLK-1 play an important role in the development of cervical carcinoma proliferation.
