摘要
目的探讨表面外胚层条件性敲除线粒体融合蛋白2(Mfn2)基因对小鼠视网膜发育的影响。方法将选择携带Le-Cre^+/-;Mfn2^fl/fl基因的雄鼠与Mfn2^fl/fl的雌鼠交配,将子代中Le-Cre^+/-;Mfn2fl/fl(Mfn2CKO)和Mfn2^fl/fl(Mfn2WT)的小鼠分别设为实验组与对照组。通过大体解剖、免疫组织化学和HE染色,观察Mfn2CKO鼠与Mfn2WT鼠视网膜形态发育的差异。结果在胚胎期发育过程中,Mfn2基因条件性敲除小鼠视网膜各层厚度不均,视网膜细胞增殖数较对照组减少,细胞凋亡增加。Mfn2基因在视网膜中主要位于光感受器外节细胞层。结论Mfn2基因影响小鼠视网膜形态发育,并与视网膜细胞的增殖凋亡相关。
Objective To establish an Mfn2 conditional knockout(Mfn2 CKO)mice model using the Cre-loxP system and investigate the effect of surface ectoderm-specific Mfn2 CKO on the mice retinal development.Methods Mfn2^fl/fl;Le-Cre^+/-(Mfn2 CKO)and Mfn2^fl/fl(Mfn2 WT)mice were used as the experimental group and the control group,respectively.The retinal morphology of Mfn2 CKO and Mfn2 WT mice was examined by gross anatomical and immunohistochemical analyses.Results Mfn2 CKO mice exhibited an uneven thickness of each retinal layer,a reduction in the number of proliferating retinal cells,and an increase in the number of apoptotic cells.Additionally,Mfn2 was mainly localized to the outer segment of the photoreceptor.Conclusion Mfn2 plays an important role in the development of retinal morphology and is associated with the proliferation and apoptosis of retinal cells in mice.
