养心活血解毒方对病毒性心肌炎模型大鼠炎症因子、相关蛋白表达及氧化应激的影响 被引量:7

Effect of Yangxin Huoxue Jiedu Decoction on Inflammatory Factors,Related Protein Expression and Oxidative Stress in Rats with Viral Myocarditis(VMC)

摘要 目的观察养心活血解毒方对病毒性心肌炎(VMC)模型大鼠炎症因子、相关蛋白表达及氧化应激的影响。方法将40只雄性SD大鼠随机分为4组,每组10只,分别为对照组、模型组、阳性组和试验组,采用柯萨奇B3病毒诱导制作VMC大鼠模型。采用ELISA检测大鼠外周血肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、白介素-17(IL-17)和白介素-1β(IL-1β)心肌酶谱指标和肌钙蛋白Ⅰ(cTnⅠ)含量以及超氧化物歧化酶(SOD)和血清丙二醛(MDA)水平的变化,采用Western blotting检测相关蛋白Caspase-3和Caspase-9蛋白。并采用光镜观察各组大鼠心肌损伤程度。结果模型组的炎症因子、Caspase-3和Caspase-9蛋白含量、心肌酶谱指标、cTnⅠ含量以及MDA含量明显升高(P<0.05),而SOD含量明显降低(P<0.05);给予养心活血解毒方和利巴韦林治疗后大鼠炎症因子、Caspase-3和Caspase-9蛋白含量、心肌酶谱指标、cTnⅠ含量以及MDA含量明显下降(P<0.05),SOD含量明显上升(P<0.05),心肌损伤明显降低,且阳性组和试验组心肌酶谱指标和cTnⅠ含量以及SOD含量比较,差异有统计学意义(P<0.05),试验组的心肌损伤明显低于阳性组。结论养心活血解毒方可降低VMC模型大鼠的炎症因子水平,改善心肌相关蛋白表达,降低心肌损伤,抑制氧化应激反应。 Objective:To investigate the effects of Yangxin Huoxue Jiedu Decoction on inflammatory factors,related protein expression and oxidative stress in VMC rats.Methods:40 male SD rats were randomly divided into four groups,10 rats in each:the control group,the model group,the positive group and the experimental group.The VMC rat model was induced by Coxsackie B3 virus.ELISA was used to detect TNF-α,IL-6,IL-17 and IL-1βmyocardial zymogram indicators,cTnⅠcontents,SOD and MDA levels.The related proteins Caspase-3 and Caspase-9 were detected by Western blotting method.The degree of myocardial damage in each group was observed by light microscopy.Results:The inflammatory factors,Caspase-3,Caspase-9 protein contents,myocardial zymogram,cTnⅠcontents and MDA contents in the model group significantly increased(P<0.05),while SOD significantly decreased(P<0.05),and the myocardial injury was obvious;inflammatory factor,Caspase-3,caspase-9 protein contents,myocardial zymogram index,cTnⅠand MDA contents decreased significantly after treatment with Yangxin Huoxue Jiedu Decoction and ribavirin(P<0.05);myocardial enzyme index,cTnⅠcontent and SOD content increased significantly(P<0.05);myocardial damage was significantly reduced,and the myocardial zymogram index,cTnⅠcontents,SOD levels were significantly different between the positive group and the experimental group(P<0.05).The myocardial damage of the experimental group was significantly lower than that of the positive group.Conclusion:Yangxin Huoxue Jiedu Decoction can reduce inflammatory factor levels,improve the expression of myocardial related protein,reduce myocardial injury and inhibit oxidative stress response.
