摘要
目的观察肺癌细胞中TNFRSF19对增殖及锚定非依赖生长能力的影响。方法将TNFRSF19连接至pLVX-EF1A慢病毒载体中,构建过表达质粒,收取慢病毒上清;感染人肺癌细胞系A549细胞后通过Western印迹实验检测TNFRSFL9表达水平的改变;通过MTT实验、平板克隆形成及肿瘤成球实验观察TNFRSF19在肺癌细胞中对增殖及锚定非依赖生长能力的影响。结果成功构建TNFRSF19慢病毒过表达质粒,并建立TNFRSF19过表达的A549细胞系。实验结果显示过表达TNFRSF19可抑制肺癌细胞增殖及锚定非依赖生长能力。结论TNFRSF19可能是一个潜在的抑癌基因,在肺癌发展过程中可能存在保护性作用。
Objective To investigate the role of TNFRSF19 in lung cancer cell. Methods Human lung cancer A549 cell line stably over-expressing TNFRSF19 was established by using lentivirus system. The stable transfection efficiency was then verified by Western blotting. The cell proliferation and anchorage-independent growth were detected by MTT assay, colony formation assay and sphere formation assay after TNFRSF19 overexpression. Results The A549 cell line with stable 77VFRSF19 overexpression was established. The function assay of TNFRSF19 in lung cancer cells showed that TNFRSF19 suppressed cell proliferation and anchorage-independent growth. Conclusion TNFRSF19 serves as a potential tumor suppressor in the development of lung cancer.
