摘要 目的建立高效液相色谱法测定人胃癌SGC-7901细胞内新藤黄酸含量的方法.方法采用高效液相色谱法,选取人胃癌SGC-7901细胞(贴壁细胞),色谱条件:色谱柱采用COSMOSIL C18,检测波长为360 nm,流动相为乙腈-0.1%乙酸水溶液(9:1),流速1 mL/min,柱温30℃.并对线性、稳定性、精密度、加样回收率和SGC-7901细胞内样品进行考察.结果新藤黄酸在(0.5~16)μg/mL浓度范围内线性关系良好,回归方程Y=3256X+517(r^2=0.9995,n=6),稳定性实验5 d、10 d后RSD分别为1.73%、1.42%,精密度实验0.5、1.0、2.0、4.0μg/mL浓度的RSD分别为1.33%、1.75%、0.76%、1.28%,平均加样回收率为100.11%(RSD=0.57%,n=4).每毫克蛋白含药量为1.22、1.17、1.24、1.19、1.21μg/mg,RSD=2.24%.结论本实验方法简便可靠、精密度高、重复性好,适用于人胃癌SGC-7901细胞内新藤黄酸含量的测定. Objective To establish a high performance liquid chromatography(HPLC) method for the content determination of neogambogic acid in human gastric cancer SGC-7901 cells. Methods Human gastric cancer SGC-7901 cells(adherent cells) were selected by high performance liquid chromatography(HPLC). Chromatographic conditions: COSMOSIL C18 was for chromatographic column and the detection wavelength was 360 nm. The mobile phase was acetonitrile-0.1% aqueous acetic acid(9:1), the flow rate was 1 mL/min, and the column temperature was 30℃. Linearity, stability, precision, sample recovery rate and intracellular sample of SGC-7901 were investigated. Results The neogambogic acid had a good linear relationship within the concentration range of(0.5-16)μg/mL. The regression equation was Y=3256X+517(r^2=0.9995, n=6). The RSD of the stability experiment was 1.73% and 1.42% after 5 d and 10 d, respectively. The RSD of the precision experiments at 0.5, 1.0, 2.0, and 4.0 μg/mL were 1.33%, 1.75%, 0.76%, and 1.28%, respectively. The average sample recovery rate was 100.11%(RSD=0.57%, n=4). The amount of protein per mg was 1.22, 1.17, 1.24, 1.19, 1.21 μg/mg, RSD=2.24%. Conclusion This experimental method is simple, reliable, and with high precision and good repeatability. It is suitable for the content determination of neogambogic acid in human gastric cancer SGC-7901 cells.
