摘要
目的:探讨有氧运动训练对Apelin基因敲除鼠葡萄糖耐量和骨骼肌糖代谢相关基因表达变化的影响。方法:8周龄野生C57BL/6J小鼠和Apelin敲除小鼠各40只,两种鼠进一步分为安静组和运动组,每组20只,共4组,分别为:野生安静组、野生运动组、敲除安静组、敲除运动组。运动组进行前两周跑速为15 m/min、后两周跑速为20 m/min、坡度为5°的跑台运动,每周运动6天,每天运动1小时。运动组最后一次运动休息48 h后,4组中每组10只进行糖耐量实验(GTT);每组剩余的10只,进行骨骼肌指标的测定。RT-PCR测量骨骼肌Apelin及糖代谢相关基因Glut4、Gbe1、Phka1、Hk2和Pfkm mRNA表达量;Western blot法测定骨骼肌Apelin和Glut4蛋白含量。结果:(1)敲除安静组与野生安静组相比,GTT曲线下面积(AUC)显著增加(P<0.01),骨骼肌内Glut4,Gbe1,Phka1,Hk2和Pfkm mRNA和Glut4蛋白表达都显著性降低(P<0.05)。(2)敲除运动组和野生运动组相比,骨骼肌内Glut4,Gbe1,Hk2和Pfkm mRNA和Glut4蛋白的表达显著性降低(P<0.05)。敲除运动组和敲除安静组相比,小鼠GTT显著性改善,骨骼肌内Glut4,Gbe1,Phka1和Hk2 mRNA表达显著性增加(P<0.05)。(3)野生运动组和野生安静组相比,小鼠糖耐量和骨骼肌内Apelin mRNA和蛋白以及糖代谢相关基因表达没有变化。结论:(1)Apelin KO鼠的糖耐量受损,骨骼肌葡萄糖代谢相关基因的表达降低;(2)4周有氧运动可改善Apelin敲除鼠的糖耐量降低,对骨骼肌糖代谢相关基因表达有一定的积极促进作用。
Objective To investigate the effect of conducting aerobic exercise on the glucose tolerance and expressions of genes related to glucose metabolism of the apelin knockout(KO) mice. Methods Forty eight-week wild C57BL/6 J mice and apelin knockout mice were randomly divided into a wild control group,a wild exercise group,a knockout control group,and a knockout exercise group,each of 20. Those in exercise groups underwent a 60-minute treadmill running six days a week for 4 weeks. The speed was at 15 m/min on a slope of 5° in the first two weeks,and then adjusted to 20 m/min in the next two weeks. The glucose tolerance test(GTT) was conducted for a half mice of each groups 48 hours after last running,while for the other half,the mRNA expressions of apelin,glucose transporter 4(Glut4),glycogen-branching enzyme(Gbe1),phosphorylase kinase,alpha 1(Phka1),hexokinase 2(Hk2) and phosphofructokinase,muscle(Pfkm) in the skeletal muscle were determined using the reverse transcription-polymerase chain reaction(RT-PCR). The content of Apelin and Glut4 protein in the skeletal muscle was measured using the Western blotting. Results Compared with the wild control group,the area under the GTT curve(AUC) increased significantly,while the expressions of Glut4,Gbe1,Phka1,Hk2 and Pfkm mRNA and Glut4 protein decreased significantly in the knockout control group(P<0.05). The expressions of Glut4,Gbe1,Hk2 and Pfkm mRNA and Glut4 protein of the knockout exercise group were significantly lower than those of the wild exercise group(P<0.05). After 4 weeks of intervention,the AUC of GTT of the knockout exercise group was significantly higher than the knockout control group(P<0.05). Compared with the knockout control group,the expressions of Glut4, Gbe1, Phka1 and Hk2 mRNA increased significantly in the knockout exercise group(P<0.05). Between the wild-type exercise group and the wild-type control group,there were no significant differences in the glucose tolerance and expressions of proteins and genes related to glucose metabolism in the skeletal muscle. Conclusion Apelin KO mice show impaired glucose tolerance and reduced expression of genes related to the glucose metabolism in skeletal muscles. The 4-week aerobic exercise improves the glucose tolerance of apelin knockout mice,and promotes the expression of genes related to the glycogen metabolism in the skeletal muscle.
