摘要
目的:研究五味子乙素对抑郁模型大鼠海马神经递质含量、炎症及氧化应激程度的影响。方法:选择雄性SD大鼠并随机分为对照组、模型组、五味子乙素组,后两组按照慢性轻度不可预见性应激建立抑郁模型,五味子乙素组在造模后给予五味子乙素灌胃,持续6周。比较3组间抑郁行为及海马在神经递质、炎症指标、氧化应激指标的差异。结果:与对照组比较,模型组大鼠糖水偏爱实验的糖水偏爱分数明显降低,强迫游泳实验的不动时间明显延长,海马中5-羟色胺(5-HT)、去甲肾上腺素(NE)、多巴胺(DA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)的含量明显减少,海马中丙二醛(MDA)的含量及核因子-κB(NF-κB)、白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的表达量明显增多(P<0.05);与模型组比较,五味子乙素组大鼠的糖水偏爱分数明显增加,不动时间明显缩短,海马中5-HT、NE、DA、SOD、GPx的含量明显增多,海马中MDA、NF-κB、IL-1β、TNF-α的表达量明显减少(P<0.05)。结论:五味子乙素能够改善抑郁模型大鼠的抑郁行为并增加单胺类神经递质分泌、抑制炎症及氧化应激反应。
Objective: To study the effects of schisandrin B on neurotransmitter content, inflammation and oxidative stress in hippocampus of depression model rats. Methods: Male SD rats were randomly divided into control group, model group and schisandrin B group. The latter two groups were established depression model according to chronic mild unpredictable stress. Schisandrin B group was given schisandrin Bgavage for 6 weeks. The differences of depressive behavior and hippocampal neurotransmitter, inflammatory index, oxidative stress index were compared among the three groups. Results: Compared with the control group, the sugar preference score was significantly lower, the immobility time was significantly prolonged, the contents of 5-hydroxytryptamine(5-HT), norepinephrine(NE), dopamine(DA), superoxide dismutase(SOD), glutathione peroxidase(GPx) in hippocampus were significantly decreased, and the contents of malondialdehyde(MDA), the expression of nuclear factor-kappa B(NF-κB), interleukin-1 beta(IL-1β) and tumor necrosis factor-α(TNF-α) in hippocampus were significantly increased in the model group(P<0.05);compared with the model group, the sugar preference score was significantly increased, immobility time significantly shortened, the contents of 5-HT, NE, DA, SOD and GPx in hippocampus were significantly increased, and the content of MDA and the expression of NF-κB, IL-1β and TNF-α in hippocampus were significantly decreased in ketamine group(P<0.05). Conclusion: Low dose ketamine can improve depressive behavior, increase monoamine neurotransmitter secretion, inhibit inflammation and oxidative stress in depression model rats.
