胃腺癌组织eIF3a和MMP-12表达的研究

摘要 目的检测胃腺癌组织真核翻译起始因子3a(eIF3a)和基质金属蛋白酶-12(MMP-12)表达,分析其临床价值。方法选择83例胃腺癌患者作为观察1组,留取临床资料及术后组织,在83例中选择39例距肿物边缘>5cm正常胃黏膜组织作为对照1组,术后留取组织石蜡包埋。在上述病例中分别选择12例胃腺癌新鲜组织作为观察2组,12例新鲜正常胃黏膜组织作为对照2组。应用免疫组化法检测观察1组和对照1组eIF3a和MMP-12蛋白表达阳性率,应用Western Bolt法检测观察2组和对照2组eIF3a和MMP-12表达量。结果免疫组化法显示,观察1组和对照1组eIF3a和MMP-12表达阳性率差异有统计学意义(eIF3a:49.40%比7.70%,P<0.05;MMP-12:42.17%比5.13%,P<0.05)。观察1组e IF3a和MMP-12表达与淋巴结转移和脉管累犯相关(P<0.05),eIF3a表达与Ki67指数和肿瘤最大径相关(P<0.05),MMP-12表达与间质炎细胞浸润程度相关(P<0.05)。观察1组e IF3a和MMP-12表达具有正相关性(r=0.46,P=0.0409),观察1组二者的表达与生存时间相关(P<0.05)。Western Bolt法显示,观察2组和对照2组eIF3a和MMP-12表达量差异有统计学意义[eIF3a:(1.21±0.21)比(0.91±0.15),P<0.05;MMP-12:(1.03±0.27)比(0.59±0.16),P<0.05]。结论胃腺癌组织eIF3a和MMP-12蛋白表达上调,且二者具有正相关性,可能共同促进病变的形成和进展。 Objective To detect the expression of Eukaryotic Translation Initiation Factor 3 a(eIF3 a) and Matrix Metalloproteinase-12(MMP-12) in gastric adenocarcinoma tissues and to analyze their clinical value. Methods 83 patients with gastric adenocarcinoma were recruited as observation group 1, clinical data and postoperative tissues were collected. 39 normal gastric mucosa tissues more than 5 cm from the edge of the tumor were defined as control group 1, and paraffin-embedded tissues were collected after operation. Among the above cases, 12 fresh gastric adenocarcinoma tissues were selected as observation group 2 and 12 fresh normal gastric mucosa tissues as control group 2, respectively. The expression of eIF3 a and MMP-12 in group 1 and control group 1 was detected by immunohistochemistry, and the expression of eIF3 a and MMP-12 in group 2 and control group 2 was tested by Western blotting. Results Immunohistochemical staining showed that the positive rates of eIF3 a and MMP-12 in group 1 and control group 1 were significantly different(eIF3 a: 49.40% vs 7.70%, P<0.05;MMP-12: 42.17% vs5.13%, P<0.05). The expression of eIF3 a and MMP-12 was correlated with lymph node metastasis and vascular involvement(P<0.05), eIF3 a expression was correlated with Ki67 index and maximum diameter of tumors(P<0.05),and MMP-12 expression was correlated with the degree of infiltration of interstitial inflammatory cells(P<0.05). The expression of eIF3 a and MMP-12 in group 1 was positively correlated(r=0.46, P=0.0409), and the expression of eIF3 a and MMP-12 in group 1 was correlated with survival time(P<0.05). The results of Western blot showed that there were significant differences in the expression of eIF3 a and MMP-12 between the two groups[eIF3 a:(1.21±0.21)vs(0.91±0.15), P<0.05;MMP-12:(1.03±0.27) vs(0.59±0.16), P<0.05]. Conclusion The expression of eIF3 a and MMP-12 was up-regulated in gastric adenocarcinoma tissues, and there was a positive correlation between them, which may promote the formation and progress of gastric adenocarcinoma.
