摘要
目的观察半夏泻心汤对重症急性胰腺炎(SAP)大鼠肠道损伤的保护作用及肠组织p38MAPK/核转录因子-κB(NF-κB)表达的影响并探讨其机制。方法将雄性8周龄SD大鼠30只均分为假手术组、模型组和半夏泻心汤组,用牛磺胆酸钠建立SAP模型,造模2 h后半夏泻心汤组以5 g/kg进行灌胃。24 h后检测血清中血清淀粉酶(AMY)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量,观察大鼠胰腺和回肠的病理改变,检测p38MAPK、p-p38MAPK及NF-κB在回肠组织中的表达情况。结果实验结束时模型组和半夏泻心汤组分别死了3只和1只大鼠;与假手术组比较,模型组和半夏泻心汤组AMY、IL-6、IL-1β和TNF-α含量增高;与模型组比较,半夏泻心汤组AMY、IL-6、IL-1β和TNF-α含量降低;假手术组胰腺和回肠未见病理损害,模型组胰腺组织出现明显水肿、出血、脂肪组织坏死及炎细胞浸润,回肠组织绒毛上皮脱落,仅有固有膜,同时伴有水肿炎细胞浸润,胰腺和回肠病理评分较假手术组增加;给予半夏泻心汤治疗后,胰腺和回肠的病理损伤较模型组明显减轻,病理学评分也降低;模型组p38MAPK、p-p38MAPK及NF-κB表达均较假手术组增加;半夏泻心汤组p38MAPK、p-p38MAPK及NF-κB表达较模型组减少,但较假手术组增加(均P <0.05)。结论半夏泻心汤通过减少p38MAPK/NF-κB表达从而发挥SAP大鼠肠道保护作用。
Objective:To investigate the protective effect of Banxia Xiexin Decoction on intestinal injury of rats with severe acute pancreatitis(SAP)and its effect on expression of p38MAPK/NF-κB in intestinal tissue. Methods:30 male 8-week-old SD rats were divided into the sham operation group,the model group and Banxia Xiexin Decoction group. SAP model was induced by the sodium taurocholate. After 2 hours of modeling,Banxia Xiexin Decoction group was administered with 5 g/kg Banxia Xiexin Decoction. The serum levels of AMY,IL-6,IL-1βand TNF-α were detected 24 h later. The pathological changes of pancreas and ileum were observed. And the expression levels of p38MAPK,p-p38MAPK and NF-κB in ileum were detected. Results:At the end of the experiment,3 rats in the model group and 1 rat in Banxia Xiexin Decoction group died. Compared with the sham-operated group,AMY,IL-6,IL-1β and TNF-α contents in the model group and Banxia Xiexin Decoction group increased. Compared with the model group,those in Banxia Xiexin Decoction group decreased. There were no pathological changes in pancreas and ileum in the sham operation group. In the model group,there were obvious edema,hemorrhage,necrosis of adipose tissue and infiltration of inflammatory cells in pancreatic tissue. The villous epithelium of ileum was exfoliated with only lamina propria,accompanied by infiltration of edematous inflammatory cells.The pathological score of pancreas and ileum was higher than that of the sham operation group. After treatment,the pathological damage of pancreas and ileum was significantly reduced and the pathological score was also reduced. The expressions of p38MAPK,p-p38MAPK and NF-κB in the model group were higher than those in the sham-operated group,while those in Banxia Xiexin Decoction group were lower than those in the model group,but higher than those in the sham-operated group. Conclusion:Banxia Xiexin Decoction can protect intestinal tract of SAP rats by reducing the expression of p38MAPK/NF-κB.
