摘要
目的分析miR-378在宫颈癌患者组织、外周血中的表达特征及临床意义。方法收集2017年3月-2018年3月本院收治的宫颈癌患者40例(CC组)、宫颈上皮内瘤变患者37例(CIN组)、对照组40例作为研究对象,q-RTPCR分析miR-378在宫颈组织标本和外周血中的表达差异,并绘制受试者工作特征曲线(ROC),分析外周血miR-378的诊断效能。结果与对照者相比,miR-378在宫颈病变患者(CC及CIN)的宫颈组织及外周血中表达上调,差异有统计学意义(P均<0. 01);根据有无淋巴结转移,将CC组分为无淋巴结转移组LN(-)和有淋巴结转移LN(+),相比于LN(-),miR-378在LN(+)的宫颈组织及外周血中表达上调,差异均有统计学意义(P <0. 01);ROC曲线分析显示,外周血中miR-378对宫颈病变具有较强的诊断能力,曲线下面积为0. 924,敏感度为81. 80%,特异性为95. 00%。结论 miR-378在宫颈组织及外周血中的高表达可能促进了宫颈病变,提示miR-378有望作为新型分子标志物应用于宫颈病变的早期诊断。
Objective To analyze the characteristic expression and clinical significance of miR-378 in tissues and peripheral blood of patients with cervical cancer. Methods 40 patients with cervical cancer( CC group),37 patients with cervical intraepithelial neoplasia( CIN group) and 40 healthy controls in our hospital were enrolled in this study from March 2017 to March 2018. The expressions of miR-378 in cervical tissue samples and peripheral blood were analyzed by q-RT-PCR,and the receiver operating characteristic( ROC) curve was performed to analyze the diagnostic efficacy by miR-378 expression in peripheral blood. Results Compared with control group,miR-378 was up-regulated in cervical tissues and peripheral tissues of patients with cervical lesions( CC and CIN)( P < 0. 01);According to the presence or absence of lymph node metastasis( LN),the CC group was divided into no lymph node metastasis group LN(-) and lymph node metastasis LN(+).Compared with LN(-),the expressions of miR-378 in both cervical tissues and peripheral blood were significantly up-regulated in LN(+),with the differences statistically significant( P < 0. 01);ROC curve analysis showed that miR-378 in the peripheral blood had strong diagnostic ability for cervical lesions. The area under the curve was 0. 924,sensitivity was 81. 80% and specificity was 95. 00%. Conclusion High expression of miR-378 in cervical tissue and peripheral blood may promote cervical lesions,suggesting that miR-378 is expected to be used as a novel molecular marker for early diagnosis of cervical lesions.
