摘要
目的:研究雄黄对人乳腺癌细胞MCF-7增殖及凋亡的影响。方法:采用MTT法检测不同质量浓度的雄黄对人乳腺癌细胞MCF-7增殖的影响;Hoechst 33258染色法观察药物作用MCF-7细胞后细胞形态的变化;流式细胞术检测药物对MCF-7细胞周期和凋亡率的影响。结果:与空白对照组相比,雄黄可显著抑制MCF-7细胞增殖(P<0.01),且呈现浓度和时间依赖性;Hoechst 33258染色和Annexin V-FITC/PI双染法结果显示,80和160μg·ml^-1雄黄处理MCF-7细胞24 h后能有效诱导其凋亡(P<0.05),且随药物浓度的增加,凋亡细胞明显增多;PI单染法检测周期结果显示,不同质量浓度雄黄处理MCF-7细胞24 h,与空白对照组比较,给药组表现为G0/G1期细胞减少,G2/M期细胞则显著增加,其中各药物组G0/G1期、G2/M期与空白对照组比较,差异有统计学意义(P<0.05)。结论:雄黄可显著抑制人乳腺癌细胞MCF-7的增殖,同时可诱导其凋亡和G2/M期阻滞。
Objective: To investigate the effects of Realgar on the proliferation and apoptosis of human breast cancer cell line MCF-7.Methods: MCF-7 cells were treated with different concentrations of Realgar,and MTT method was used to evaluate the cell proliferation.The morphology of MCF-7 cells with Hoechst33258 staining was observed under a fluorescent microscope.Flow cytometry was used to analyze the cell cycle and the apoptosis of MCF-7 cells.Results: Compared with the blank control group,Realgar could markedly inhibit the proliferation of MCF-7 cells in a dosage-and time-dependent manner (P<0.01);Hoechst33258 fluorochrome staining and Annexin V-FITC/PI double staining showed that different concentrations of Realgar (80 and 160 μg·ml^-1)could promote MCF-7 cells apoptosis (P<0.05),the apoptosis rate was gradually increased with the drug concentration increase;propidium iodide (PI) staining method indicated that there were less cells in G0 /G1 phase and more cells in G2 /M phase when compared with the blank control group and the above differences were statistically significant (P<0.05).Conclusion: Realgar can significantly inhibit the proliferation of human breast cancer MCF-7 cells,and induce the cell apoptosis and the arrest of G2 /M phase.
