狐肺炎大肠杆菌ybjX基因缺失株构建及其对小鼠致病性检测

Construction and pathogenicity assay of a YbjX-deficient mutant of E.coli HBCLE-12 strain causing pneumonia in foxes with mouse model

摘要 为研究外膜蛋白YbjX对大肠杆菌致病作用的影响,本研究利用λ-Red同源重组系统构建了狐肺炎大肠杆菌(HBCLE-12)ybjX基因缺失突变株(HBCLEΔybjX),并对其生物学特性进行分析,探索YbjX蛋白在大肠杆菌致病过程中的作用。结果表明,缺失株HBCLEΔybjX与野生型菌株和回补菌株相比,其生长速度、生化特性无明显差异,但其运动能力和血清内存活能力显著下降,对多种抗生素的敏感性明显增强。动物试验表明,ybjX基因缺失突变株对小鼠的致病力显著下降。本研究为进一步阐释大肠杆菌致病机制和疫苗设计奠定基础。 To investigate the roles of outer membrane protein(Omp)YbjX in pathogenic processes of E.coli.λ-Red homologous recombination was used for knockout of ybjX gene,and comparison of growth characteristics,biochemical characteristics and bacterial virulence were performed for analysis the change between the HBCLE-12wild type strain and ybjX knockout mutant.The data demonstrated that,the YbjX-deficient mutant showed no differences in growth characteristics and biochemical characteristics with wild type strains and complemented strains.But compared with the wild type and complemented strains,the mutant displayed sharply decreased survival rate in both foxes and mouse serum,attenuated migration in 0.25%agar,and significantly increased sensitivity to multiple antibotics.The apparently attenuated virulence was also observed in YbjX-inactivation mutant.The experiment generated a YbjX-deficient mutant of E.coli,providing foundation for exploring underlying pathogenic mechanism of E.coli and vaccine design.
