摘要
目的通过检测哮喘肺组织Bid和骨膜蛋白的表达水平,探讨Bid和骨膜蛋白参与哮喘炎症的机制。方法复制哮喘大鼠模型并取肺组织,采用免疫组化法和实时定量PCR法检测肺组织骨膜蛋白、BidmRNA与Bid蛋白的表达。结果哮喘组骨膜蛋白、BidmRNA和Bid蛋白的表达水平高于对照组(均P<0.05)。肺组织BidmRNA与Bid蛋白间、Bid蛋白与骨膜蛋白间均呈正相关(r=0.61、0.60,均P<0.05)。结论哮喘肺组织中骨膜蛋白和Bid可能具有促进哮喘气道炎症的作用。
Objective To investigate the potential roles of Bid and periostin in asthmatic inflammation pathogenesis by detecting the expression level of Bid and periostein in asthma lung tissue.Methods Rat asthma model were replicated and lung tissuewas obtained, and then the expression levels of periostin, BidmRNA and Bid protein were detected by immunohistochemisty or real-time polymerase chainreactionmethods at lung tissue.Results The expression levels of periostin, Bid mRNA and Bid proteins in asthma group were higher than those in control group (all P< 0.05).Moreover, therewas significantly positive correlation either between levels of BidmRNA and Bid protein or between levels of Bid protein and periostin protein (r=0.61,0.60, all P< 0.05).Conclusions The expression level of periostin and Bid in asthmatic lung tissue increased, and they may have the effect of promoting airway inflammation in asthma.
