解淀粉芽孢杆菌G9R-3脂肽类化合物抑制香蕉枯萎病菌机理及防效评价 被引量:7

摘要 [目的]G9R-3是从抗病香蕉品种“桂蕉9号”健康植株根部中分离得到的一株具有较强抑制香蕉枯萎病菌(Fusarium oxysporum f.sp.cubense race 4,Foc4)活性的内生解淀粉芽孢杆菌(Bacillus amyloliquefaciens)。开展其脂肽类化合物抑制香蕉枯萎病菌活性机理及防效评价研究,为利用该菌株大田防控香蕉枯萎病提供参考。[方法]结合基质辅助激光解吸电离-飞行时间质谱(MALDI-TOF-MS)和液相色谱-质谱联用(LC-MS)分析脂肽类化合物类型,采用Landy培养基发酵并使用大孔树脂抽提法提取菌株脂肽类物质,结合室内平板、台盼蓝染色、盆栽灌根接种进行活性检测与防效评价。[结果]该菌株产生的脂肽类化合物种类包括表面活性素Surfactin、泛革素Fengycin和伊枯草菌素iturins 3个种类。活性检测表明,当粗提物浓度为300μg/mL时对香蕉枯萎病菌Foc4菌丝生长抑制率达58.91%,孢子萌发抑制率达69.80%。室内盆栽防效评价试验表明,浓度为1 mg·mL-1 G9R-3脂肽类粗提物香蕉枯萎病防效可达76.53%。[结论]G9R-3产生的脂肽类化合物具有防控香蕉枯萎病的潜力,其生防基质为破坏Foc4细胞膜并引起菌丝肿大和畸形,并抑制其孢子萌发。 [Objective]An endophytic Bacillus amyloliquefaciens strain,G9 R-3 possessed antifungal activity from Disease-resistant banana variety k Guijiao 9 '.To provide reference for field control of banana fusarium wilt,the mechanism research and control effects of its lipopeptide compounds against banana fusarium wilt were carried out.[Method]lipopeptides were identified from cultural of G9 R-3 by using MALDITOF-MS and LC-MS.Landy medium was used for large scale fermentation and macroporous resin extraction was used to extract lipopeptides.The activity detection and control effect evaluation were carried out in combination with indoor plate,trypan staining assay and potted root irrigation.[Result]The strain can produce surfactin,fengycin and iturins.Antifungal analysis result indicated that when concentration of lipopeptide extraction reached 300μg/mL,the inhibition rate of hyphae growth in FOC4 of banana fusarium wilt could reach58.91%,the spore germination was 69.80%.The indoor potted effect evaluation test indicated that the concentration of 1 mg · mL^(-1)G9 R-3 lipopeptides crude extract effect could reach76.53%.[Conclution]Lipopeptide extraction from G9 R-3 culture could destroy cell membrane of Foc4,cause hyphae enlargement and deformity as well as inhibit spore germination.
