摘要
目的:利用超高效液相色谱-二极管阵列检测法(UPLC-PDA)同时检测加味四君子汤水煎液中6种主要抗癌成分的含量。方法:采用WatersBEH(R)C18色谱柱(100mm×2.1mm,1.7μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,柱温为25℃,流速为0.2mL/min。结果:各对照品在线性范围内线性关系良好,r≥0.999,6;加样回收率为98.86%~101.86%,RSD均≤3.45%。加味四君子汤水煎液中人参皂苷Re、白术内酯Ⅲ、茯苓酸、甘草酸、齐墩果酸和小檗碱的含量分别为70.15~71.32、12.67~14.03、17.77~34.34、98.72~103.89、31.37~38.73、13.00~13.20mg/L。结论:该方法操作简便,各峰的分离度好,结果准确,重复性高,能够高效检测加味四君子汤中6种抗癌成分含量,可作为加味四君子汤的质控方法,为进一步研究提供参考。
Objective:To determine the content of six main anti-cancer components in Modified Sijunzi Tang by UPLC-PDA simultaneously.Methods:The test was performed with Waters BEN(R)C18 chromatographic column(100 mm×2.1 mm,1.7μm),and using acetonitrile-0.1%phosphoric acid solution as mobile phase for gradient elution.The column temperature was 25℃and flow rate at 0.2 mL/min.Results:All controls had good linear relations within the linear range with r≥0.999,6.The recovery rate was 98.86%-101.86%,and the RSD≤3.45%.Detected content of ginsenoside Re,arbutinⅢ,pachymic acid,glycyrrhizin,oleanolic acid and berberine were 70.15-71.32,12.67-14.03,17.77-34.34,98.72-103.89,31.37-38.73,13.00-13.20 mg/L respectively.Conclusion:This method is simple to operate,accurate,repeatable,and each peaks was separated.It can be used as a quality control method for flavoring Modified Sijunzi Tang and foundation of further researches.
