摘要
乳腺癌是世界上发病率最高的癌症之一,在临床治疗中以手术为主,化学药物治疗为辅.为解决化疗进行中,病患身上出现的对药物的耐受性问题,通过构建乳腺癌耐药细胞株研究OLA1变化对乳腺癌耐药性影响,依靠siRNA转染改变OLA1在细胞内的表达情况,接着使用流式细胞仪检测OLA1变化对细胞凋亡影响.最终发现OLA1通过影响细胞凋亡相关蛋白表达,降低紫杉醇对乳腺癌细胞的杀伤效果,由此提高乳腺癌细胞对药物的耐受性.
Breast cancer is one of the most common cancers in the world.Surgical resection is the main method in clinical treatment and taking medicine is a supplement.With the progress of chemotherapy,drug tolerance has become an urgent problem in clinical treatment.To solve this problem,a breast cancer drug-resistant cell line is constructed to study the effect of OLA1 changes on breast cancer drug resistance,and the expression of OLA1 in cells is changed by siRNA transfection.Then the flow cytometry is used to detect the effect of OLA1 changes on cell apoptosis.Finally,it is found that OLA1 can reduce the lethal effect of paclitaxel on breast cancer cells by affecting the expression of apoptosis-related proteins,thereby improve the drug tolerance of breast cancer cells.
