摘要
目的探讨天麻钩藤饮对自发性高血压大鼠血压和血管内皮损伤修复的影响。方法将50只自发性高血压大鼠分为模型组、西药组[依那普利3mg/(kg·d)]、中药低剂量组[1.0mL/(100g·d)]、中剂量组[1.5mL/(100g·d)]和高剂量组[2.0mL/(100g·d)],另设对照组。各组大鼠均灌胃6周,每周测量血压,末次给药后检测外周血内皮细胞生长因子(VEGF)、肿瘤坏死因子α(TNF-α)含量,并对外周血内皮祖细胞进行流式细胞计数。结果与模型组比较,中药高剂量组及西药组大鼠血压降低(P<0.05);中药低剂量组、中剂量组、高剂量组及西药组血清VEGF、TNF-α浓度较模型组均降低,差异均有统计学意义(P<0.01);中药高剂量组及西药组外周血内皮祖细胞数量升高(P<0.05)。结论天麻钩藤饮可降低自发性高血压大鼠血压,其作用机制可能与降低血管内皮细胞损伤、促进内皮祖细胞动员有关。
Objective To investigate the influence of Tianma Gouteng Decoction on blood pressure and vascular endothelial injury repair in spontaneous hypertension rats.Methods Fifty spontaneous hypertension rats were divided into model group,western medicine group[enalapril 3 mg/(kg·d)],low dose group[1.0 mL/(100 g·d)],medium dose group[1.5 mL/(100 g·d)],and high dose group[2.0 mL/(100 g·d)],and control group.Blood pressure was measured weekly after gavage for 6 weeks in each group.After the last dose of administration,the contents of endothelial cell growth factor(VEGF)and tumor necrosis factorα(TNF-α)in peripheral blood were detected,and flow cytometry of peripheral blood endothelial progenitor cells was Performed.Results Compared with the model group,the blood pressure of the spontaneous hypertension rats in the high dose group,and the western medicine group decreased(P<0.05).VEGF and TNF-αin the low dose group,the medium dose group,the high dose group and the western medicine group were lower than those in the model group(P<0.01).The number of endothelial progenitor cells in peripheral blood increased in the high dose group and the western medicine group(P<0.05).Conclusion Tianma Gouteng Decoction could reduce blood pressure in spontaneous hypertension rats,and its mechanism might be related to reducing vascular endothelial cell injury and promoting endothelial progenitor cell mobilization.
