摘要
目的探讨直肠癌组织中染色体浓缩调节因子1(RCC1)的表达及临床意义。方法采用GEPIA数据库分析RCC1在结直肠癌组织中的表达情况及与预后的关系。收集本院2015年1月至2018年10月经病理确诊的66对直肠癌组织和配对癌旁组织,采用实时定量PCR(QPCR)检测上述组织中RCC1 mRNA水平,分析RCC1表达与直肠癌临床病理特征(性别、年龄、TNM分期、肿瘤大小、分化程度、浸润深度和淋巴结转移)的关系。结果GEPIA数据库分析显示结肠癌和直肠癌组织中RCC1水平均高于正常组织(P<0.05);结直肠癌全组分析显示RCC1表达与总生存期(OS)无关,但低表达的无复发生存期(RFS)优于高表达(HR=0.83,P=0.026);结肠癌亚组分析显示低表达的OS和RFS均优于高表达(HR=0.59,P=0.034;HR=0.55,P=0.016),而直肠癌亚组分析显示RCC1表达与OS和RFS均无关(P>0.05)。直肠癌组织中RCC1 mRNA水平为1.987±0.713,高于癌旁组织的1.152±0.561(P<0.05)。RCC1表达与性别、年龄和分化程度均无关(P>0.05),而与肿瘤大小、浸润深度、淋巴结转移和TNM分期均有关(P<0.05)。结论肿瘤细胞RCC1的异常表达可能参与人直肠癌的发生发展,RCC1表达可能是肿瘤细胞进展和预后不良的潜在标志物。
Objective To investigate the expression of regulator of chromosome condensation 1(RCC1)in rectal cancer tissues and its clinical significance.Methods GEPIA database was used to analyze the expression of RCC1 in colorectal cancer tissues and its relationship with prognosis.Sixty-six pairs of rectal cancer tissues and matched normal tissues confirmed by pathology from January 2015 to October 2018 were collected in our hospital.The mRNA levels of RCC1 in the above tissues were detected by real-time quantitative PCR(QPCR).The relationship between RCC1 expression and the clinicopathological parameters of rectal cancer(sex,age,TNM stage,tumor size,differentiation,depth of invasion and lymph node metastasis)were analyzed.Results RCC1 levels in colon cancer and rectal cancer tissues were higher than those in normal tissues by GEPIA database analysis(P<0.05).Total analysis of colorectal cancer showed that RCC1 expression was not related to overall survival(OS),but relapse-free survival(RFS)with low expression was better than those with high expression(HR=0.83,P=0.026).Subgroup analysis of colon cancer showed that both OS and RFS with low expression were better than those with high expression(HR=0.59,P=0.034;HR=0.55,P=0.016),but RCC1 expression was not related with OS and RFS of rectal cancer(P>0.05).The mRNA level of RCC1 in rectal cancer tissues was 1.987±0.713,higher than 1.152±0.561 in adjacent tissues(t=7.478,P<0.001).The expression of RCC1 was not related to sex,age and differentiation(P>0.05),but related to tumor size,depth of invasion,lymph node metastasis and TNM stage(P<0.05).Conclusion Abnormal expression of RCC1 in cancer cells may be involved in the development of human rectal cancer,and RCC1 expression may be a potential marker of poor progression and prognosis of cancer cells.
