摘要
采用微孔板法研究不同温度、培养基、NaCl质量分数和pH对非致病性和致病性副溶血弧菌成膜性的影响,以及苯扎氯铵、二氧化氯、过氧乙酸、过氧化氢、二氯异氰尿酸钠和次氯酸钠6种消毒剂对其生物膜的清除作用,并用扫描电镜观察消毒前后生物膜的变化。结果表明,副溶血弧菌在22℃、含3%NaCl的TSB培养基中的成膜性最好,最适NaCl质量分数为1.5%~5.5%,最适pH为7~8。6种消毒剂中,二氯异氰尿酸钠与次氯酸钠能显著清除生物膜。50 mg/L二氯异氰尿酸钠作用10 min与30 mg/L次氯酸钠作用5 min,对E17和156FR生物膜的清除率分别为90.22%、93.14%、92.20%和92.31%。扫描电镜下观察到,30 mg/L次氯酸钠作用20 min后生物膜被清除,菌体死亡。次氯酸钠为清除副溶血弧菌生物膜最有效消毒剂。
Microtiter-plate test method was used to evaluate the biofilm formation of non-pathogenic and pathogenic Vibrio parahaemolyticus strains with different temperatures, culture medium, NaCl concentrations and pH. The effects of six disinfectants, including benzalkonium chloride, chlorine dioxide, peracetic acid, hydrogen peroxide, sodium dichloroisocyanurate and sodium hypochlorite, on cleaning the biofilm were assessed. The change of biofilm before and after disinfection were observed by scanning electron microscope. The results showed that Vibrio parahaemolyticus had the best film-forming ability in TSB medium containing 3% NaCl at 22 ℃. The optimum NaCl concentration was ranging from 1.5% to 5.5%, and the optimum pH was 7 to 8. Among the six disinfectants, sodium dichloroisocyanurate and sodium hypochlorite have the strongest ability to scavenge biofilm. The removal rates of E17 and 156 FR biofilms were 90.22%, 93.14%, 92.20% and 92.31% respectively after 10 min of 50 mg/L sodium dichloroisocyanurate and 5 min of 30 mg/L sodium hypochlorite. Scanning electron microscopy(SEM) showed that the biofilm was cleared and the bacteria died after 20 min of sodium hypochlorite treated at 30 mg/L. Therefore, sodium hypochlorite is the most effective disinfectant for removing Vibrio parahaemolyticus biofilm.
