摘要
采用不同浓度梯度的中性红、龙胆紫及尼罗蓝染液对长白公猪精子染色,根据染色现象及精子活率,探究3种染料较为理想的作用时间和浓度。结果:0.01%中性红染色2 min后活精子着红色,死亡精子不着色;0.05%龙胆紫染色1 min后活精子着淡蓝色,死亡精子深紫色;0.1%尼罗蓝染色2 min后活精子着淡蓝色,死亡精子不着色。3种情况下精子的存活率与空白对照组比较无显著差异(P>0.05)。试验表明,以上3种染色条件能清晰地区分存活精子和死亡精子,可用于测定精子活率。
In this study, neutral red, methyl violet and Nile blue were used to stain the semen samples obtained from the Changbai boar. Based on the staining results and viabilities of the boar sperms, the concentrations and ideal time points of effects of the three staining materials were explored. The results showed that In the 0.01% neutral red solution in two-minute staining, the surviving sperms were red, and the dead ones were of no color;that in the 0.05% methyl violet solution in one-minute staining, the surviving sperms were light blue, and the dead ones were mainly purple;and that in the 0.1% Nile blue solution in two-minute staining, the surviving sperms were light blue, and the dead ones were of no color. The survival rates of sperms of under the three stainings were of no significant differences from that the blank control group(P>0.05). In conclusion, the 0.01% neutral red solution in two-minute staining, the 0.05% methyl violet solution in one-minute staining, and the 0.1% Nile blue solution in two-minute staining might all be used to distinguished the surviving sperm from the dead ones, determining the viability rate of sperms of boars.
